Bacterial community dynamics in a swine wastewater anaerobic reactor revealed by 16S rDNA sequence analysis

Abstract: Anaerobic digestion is a microbiological process of converting organic wastes into digestate and biogas in the absence of oxygen. In practice, disturbance to the system (e.g., organic shock loading) may cause imbalance of the microbial community and lead to digester failure. To examine the bacterial community dynamics after a disturbance, this study simulated an organic shock loading that doubled the chemical oxygen demand (COD) loading using a 4.5L swine wastewater anaerobic completely stirred tank reactor (C… Show more

“…Library preparation. The V1 and V3 hypervariable regions of the bacterial 16S rRNA gene were amplified using the primer pair 63F (5′-CAGGCCTAACACATGCAAGTC-3′) 36 and 511R (5′-GCGGCTGCTGGCACRKAGT-3′) 37 (Eurofins Genomics GmbH, Ebersberg, Germany), added to an overhang adapter sequence tail (5′-TCGTCGGCAGCGTCAGATGTGTATAAGAGACAG-3′), yielding a PCR product of ~545 bp. The V1-V3 primer pair covers a typical region widely used for skin microbiome studies 38 , but it's also recommended for nasopharyngeal areas 39 .…”

“…To compare the microbiota composition in thermophilic (55 ° C, TAnD) and mesophilic (37 ° C, MAnD) AnD digesters, two bench-top CSTRs (completely stirred tank reactors), each with a working volume of 3 L, were set up for this study [36]. The digesters were operated at two different temperatures, mesophilic (37 ° C) and thermophilic (55 ° C), using 15 days hydraulic retention time (HRT), and 5% total solids (TS) without pH control.…”

“…Anaerobic digestion is the most widely employed method for sewage sludge treatment, and since the last years, it is an attractive technology for processing various organic wastes produced in urban, industrial, and agricultural settings (3). In this process, a large fraction of the organic matter, which is degraded by a complex community of microorganisms, is broken down into carbon dioxide (CO2) and methane (CH4).…”
