We investigated the degradation of biphenyl and identified main degradation products. Biphenyl and 2,3-dihydroxybiphenyl (2,3-DHB) was added to cultivation media to identify whole collection of degradation products of four bacterial strains isolated from long-term PCB contaminated soil (Alcaligenes xylosoxidans and Pseudomonas stutzeri) and long-term PCB contaminated sediment (Ochrobactrum anthropi and Pseudomonas veronii). Cultivation flasks were processed in different time after inoculation to determine biphenyl fission rate. Alcaligenes xylosoxidans was revealed as the most appropriate strain for bioremediation process with the highest biphenyl transformation rate. Biphenyl degradation led to the formation of benzoic acid. However, as the presence of 2-hydroxy-6-oxo-6-phenylhex-2,4-dienoic acid (HOPDA) was not confirmed, the transformation pathway common for many other bacteria is probably modified.

Background: Due to their widespread use in industrial applications in recent decades, Polychlorobiphenyls (PCBs) and heavy metals (HMs) are the most common soil contaminants worldwide, posing a risk for both ecosystems and human health.Results: a poplar-assisted bioremediation strategy has been applied for more than four years to a historically contaminated area (PCBs and HMs) in Southern Italy using the Monviso poplar clone. This clone was effective in promoting a decrease in all contaminants and an increase in soil quality in terms of organic carbon and microbial abundance. Moreover, a significant shift in the structure and functioning of the belowground microbial community was also observed when analysing both DNA and cDNA sequencing. In fact, an increase in bacterial genera belonging to Proteobacteria and able to degrade PCBs and resist HMs was observed. Moreover, the functional profiling of the microbial community predicted by PICRUSt2 made it possible to identify several genes associated with PCB transformation (e.g. bphAa, bphAb, bphB, bphC), response to HM oxidative stress (e.g. catalase, superoxide reductase, peroxidase) and HM uptake and expulsion (e.g. ABC transporters).Conclusions: This work demonstrated the effectiveness of the poplar clone used in stimulating the natural belowground microbial community to remove contaminants (phyto-assisted bioremediation) and improve the overall soil quality. It is a practical example of a nature based solution involving synergic interactions between plants and the belowground microbial community.

Assessment of Biodegradation Efficiency of Polychlorinated Biphenyls (PCBs) and Petroleum Hydrocarbons (TPH) in Soil Using Three Individual Bacterial Strains and Their Mixed Culture

Biodegradation is one of the most effective and profitable methods for the elimination of toxic polychlorinated biphenyls (PCBs) and total petroleum hydrocarbons (TPH) from the environment. In this study, aerobic degradation of the mentioned pollutants by bacterial strains Mycolicibacterium frederiksbergense IN53, Rhodococcus erythropolis IN129, and Rhodococcus sp. IN306 and mixed culture M1 developed based on those strains at 1:1:1 ratio was analyzed. The effectiveness of individual strains and of the mixed culture was assessed based on carried out respirometric tests and chromatographic analyses. The Rhodococcus sp. IN306 turned out most effective in terms of 18 PCB congeners biodegradation (54.4%). The biodegradation index was decreasing with an increasing number of chlorine atoms in a molecule. Instead, the Mycolicobacterium frederiksbergense IN53 was the best TPH degrader (37.2%). In a sterile soil, contaminated with PCBs and TPH, the highest biodegradation effectiveness was obtained using inoculation with mixed culture M1, which allowed to reduce both the PCBs (51.8%) and TPH (34.6%) content. The PCBs and TPH biodegradation capacity of the defined mixed culture M1 was verified ex-situ with prism method in a non-sterile soil polluted with aged petroleum hydrocarbons (TPH) and spent transformer oil (PCBs). After inoculation with mixed culture M1, the PCBs were reduced during 6 months by 84.5% and TPH by 70.8% as well as soil toxicity was decreased.
