Banana Peel: A Potential Substrate for Laccase Production by Aspergillus fumigatus VkJ2.4.5 in Solid-State Fermentation

Abstract: In solid-state fermentation, among various solid supports evaluated, banana peel was found to be an ideal support and resulted into higher levels of laccase (6281.4 ± 63.60 U l(-1)) along with notable levels of manganese peroxidase production (1339.0 ± 131.23 U l(-1)) by Aspergillus fumigatus VkJ2.4.5. Maximum levels of laccase was achieved under derived conditions consisting of 80% of moisture level, 6 days of incubation period, 6% inoculum level, and an aeration level of 2.5 l min(-1). A column-tray bioreact… Show more

“…Moilanen et al [16] reported that the maximum MnP activity (340 nkat/g DM) occurred after 14 days of fermentation in a solid-state reactor with an internal conveyor, but their MnP activity stayed at almost the same level at 100 g-scale in plastic jars and 4 kg-scale in bioreactor cultivation. A similar study was carried out by Vivekanand et al [32] using a bioreactor that contained four circular aluminum trays, and the maximum MnP activity (1,339.0 ± 131.23 U/l) was obtained after 6 days of SSF by Aspergillus fumigatus VkJ2.4.5. Phanerochaete chrysosporium BKM-F-1767 had also produced 1,293 U/l of MnP activity, in a fixed-bed bioreactor on the 7th day [17].…”

“…However, largescale enzyme production from white-rot fungi is limited mainly owing to the lack of suitable bioreactor configurations for fungal growth and their metabolism [27]. Although several studies on the development of innovative bioreactor systems have already been carried out [7,25,32], an ideal bioreactor for ligninolytic enzyme production from white-rot fungi has not yet been shown [6,8]. Furthermore, the high production cost is another barrier to the commercialization of these enzymes.…”

“…Banana peel was the best substrate among the other tested lignocellulosic agro-residues, poplar leaves, wheat bran, rice bran, wheat straw, and bagasse under SSB conditions for obtaining higher laccase production. The laccase activity reached to maximum as 6281.4 U L -1 under optimized conditions of banana peel culture (Vivekanand et al 2011). Our highest laccase activity was 6.97-fold higher than that of it.…”

During the fermentation of laccase, manganese peroxidase is also produced by A. fumigatus (VkJ2.4.5) under SSF conditions (Vivekanand et al 2011). Figure 4 shows the different enzymes obtained from BP using SSF.…”
