Basic fibroblast growth factor enhances PPARγ ligand‐induced adipogenesis of mesenchymal stem cells

Abstract: Mesenchymal stem cells (MSCs) are capable of differentiating into a variety of lineages, including bone, cartilage, or fat, depending on the inducing stimuli and specific growth and differentiation factors. It is widely acknowledged that basic fibroblast growth factor (bFGF) modulates chondrogenic and osteogenic differentiation of MSCs, but thorough investigations of its effects on adipogenic differentiation are lacking. In this study, we demonstrate on the cellular and molecular level that supplementation of … Show more

“…Rather, TGF-␤, which is secreted in high amounts by our MSC in vitro (6), acts as a major signal for adipogenesis (27,28). Another factor that strongly enhances adipogenic differentiation of MSC in vitro is basic fibroblast growth factor (29), which is released within the glomerulus during early anti-Thy1.1 nephritis (30). Finally, human adipocytes express Fc receptors, and lipogenesis is stimulated by Ig as efficiently as by insulin (20).…”

“…IL-6 or transferrin supplementation had no significant effect on cell doubling time or the total number of population doublings. Thus, supplementation with AA, FGF-2, and PDGF-BB resulted overall in a greater than 1,000-fold increase in total cell numbers compared with controls (for controls 2 10.05 ϭ 895 cells; AA, 2 20.27 ϭ 1,267,893 cells; FGF-2, 2 25.59 ϭ 50,542,945 cells). Furthermore, an even greater proliferation rate was observed when a combination of AA, FGF-2, and PDGF-BB was added to the culture.…”

“…2 ). Interestingly, culture of rat BM MSCs or human ASCs in the presence of FGF2 prior to adipogenic induction upregulates expression of PPAR ␥ and enhances subsequent adipogenesis ( 224,225 ). Indeed, exogenous FGF2 facilitates in vivo WAT formation by isolated human SVCs ( 226 ).…”
