Basic leucine zipper transcription factor, ATF-like (BATF) regulates epigenetically and energetically effector CD8 T-cell differentiation via Sirt1 expression

Abstract: CD8 T cells play a critical role in protection against viral infections. During effector differentiation, CD8 T cells dramatically change chromatin structure and cellular metabolism, but how energy production increases in response to these epigenetic changes is unknown. We found that loss of basic leucine zipper transcription factor, ATF-like (BATF) inhibited effector CD8 T-cell differentiation. At the late effector stage, BATF was induced by IL-12 and required for IL-12–mediated histone acetylation and surviv… Show more

“…This raises the question of whether other STAT3 cytokines elevated during the later phases of LCMV Cl13 infection, such as IL-10, IL-27 and IL-6, can also induce BATF expression in CD8 T cells. In addition, other signals, such as IL-12 and PD-1 ligation, can also induce BATF expression in CD8 T cells (Kuroda et al, 2011; Quigley et al, 2010). The relative contribution of these various pathways to the regulation of BATF expression requires further study.…”

“…BATF-deficient mice show impaired effector CD8 + T-cell differentiation, characterized by reduced expression of IFN-γ, Perforin, and T-bet, which was caused by changes in epigenetic remodeling and energy metabolism (32). It is possible that BATF and IRF4 cooperatively affect CD8 + T-cell effector differentiation either by regulation of effector proteins and TFs or by promoting changes in metabolic pathways.…”

“…Sirt1 has emerged as a critical immune regulator by suppressing T cell immunity and macrophage functions (5,7,16,43,44), providing a rationale for Sirt1 activators in autoimmune and inflammatory disease therapy. Our findings here show that Sirt1 suppresses DC production of IL-27 and IFN-␤, two antiinflammatory cytokines that negatively regulate CD4 ϩ T cell differentiation toward Th17, indicating that Sirt1 may tune or function as a balance keeper for the immune response during inflammation.…”
