Bermuda Grass as a Potential Reservoir Host forGrapevine fanleaf virus

Abstract: Grapevine fanleaf virus (GFLV) was detected in samples of Bermuda grass (BG) from Iran by reverse transcription-polymerase chain reaction (RT-PCR) using two different pairs of GFLV-specific primers, and also by enzyme-linked immunosorbent assay (ELISA) using antiserum specific for a North American isolate of the virus. RT-PCR detected GFLV in both fresh and dried BG tissues and in virus preparations purified from these plants. Cloning and sequencing of the RT-PCR products confirmed that the amplified sequences… Show more

“…The virus was detected by RT-PCR using two different pairs of GFLV specific primers and ELISA. However, the Bermuda grass expressed few or no symptoms of GFLV infection (Izadpanah et al, 2003). In addition to X. index, X. italiae has been reported to spread GFLV , but these results were not corroborated (Esmenjaud & Bouquet, 2009).…”

“…With respect to GFLV isolates from Iran, only the CP gene has so far been partially sequenced (Izadpanah et al 2003). The goal of the current study was to survey vineyards in the northwest region of Iran for GFLV and to investigate genetic variation in the MP of representative isolates.…”

“…To that end, various methods have been developed for detecting GFLV which include ELISA [7], reverse transcription polymerase chain reaction (RT-PCR) [28], RNA oligo probe capture RT-PCR [8] and IC-RT-PCR [41]. In Iran, grapes are produced at the rate of 2.1 million tons annually on about 270,000 ha [17]. The North West region of Iran shares a major part of the produce and therein grape industry has been established.…”

“…The use of 5 0 NC and GMPR1 was expected to produce a fragment of 1,995 bp encompassing 181 nts of 5 0 UTR plus HP and MP genes. The primer cp912 [17] corresponds to nucleotides of 2,936-2,959 of GFLV-F13 RNA2.…”
