Bilayer tablets are a kind of medication that effectively treats illness by combining two pharmaceuticals, either the same or different ones, in a single dosage. Physically separating active pharmaceutical ingredients (API) may prevent chemical incompatibilities, and bilayer tablets can pave the way for the creation of various drug release profiles, such as immediate and prolonged release. Analgesic and anti-inflammatory bilayer tablets are significantly different. Two medications may be sequentially released from a bi-layer tablet, making it ideal for sustained-release tablets. One layer can be immediate-release for the first dosage, while the other can be a maintenance dose. The purpose of this review is to identify problems with bilayer tablet preparation and to suggest ways to fix them. To further aid comprehension, the article goes on to list the many kinds of bilayer tablets, including single-side presses, double-side presses, and bilayer tablet displacement presses, as well as their uses, advantages, and disadvantages. In addition, the review paper discusses the several ways and procedures used to manufacture bilayer tablets, so readers may have a comprehensive understanding of these tablets. In the last paragraph, the whole essay is evaluated critically.

Bilayer tablets are novel drug delivery systems where combination of two drugs in a single unit having different release profiles can be delivered. Bilayer tablets improve patient compliance, prolong the drug(s) action and can deliver two incompatible drugs in a single formulation. Bilayer tablets have one layer of active ingredient for immediate release and a second layer for delayed release, either as a second dose or in an extended release fashion. Bilayer tablets are advancing helpful technologies to overcome the disadvantages of single-layered tablets. However, bilayer tablet technology is resource-intensive. A thorough selection of excipients and manufacturing conditions for each technical stage is also required. Patients with high blood pressure often have difficulty or are unable to regulate their BP with just a single medication. The majority of hypertension patients will need to take two or more antihypertensive medicines in order to meet treatment goals. Combinations of antihypertensive drugs from various categories have been found to be more effective than either drug alone in treating hypertension in people whose blood pressure cannot be maintained satisfactorily with monotherapy. Combining two antihypertensive medications with mutually beneficial mechanisms of action may result in much greater blood pressure lowering efficacy than any of these components alone. The goal of this review is to discuss the use of bilayer tablets to administer fixed antihypertensive drug combinations for the treatment of high blood pressure. The present research is intended for the formulation development, optimization, and in-vitro evaluation of bi-layer tablets containing Enalapril maleate, hydrochlorothiazide in the immediate release layer and Nifedipine in the sustained release layer based on GEMINEX technology. The impact of formulation variables such as combination of polymers and coating solution on the drug release was evaluated from the developed formulation. The prepared formulation blend was assessed for compressibility characteristics. The homogeneity of each API in a blend was tested by determining the %assay of individual drugs from different layers. Both the immediate and controlled release granules were formulated into bilayer tablets by the direct compression method. SEM analysis of bi-layered tablets was also performed. The mean drug content for all formulations was found to be within specified limits. The In-vitro dissolution studies can be performed at USP type II dissolution apparatus for coated tablets. The release of Enalapril maleate and hydrochlorothiazide from the immediate-release layer was found to be 99.2+0.8% and 99.5+1.1, and Nifedipine from the sustained release layer was found to be 100.7%+0.5%, respectively. Hence the bilayer tablets of Enalapril maleate Hydrochlorothiazide and Nifedipine were used to improve patient compliance towards the effective management of hypertension. INTRODUCTION:Hypertension is an unusual high arterial blood pressure. Hypertension is a major risk factor for various cardiovascular diseases (CVDs)uncontrolled hypertension increases the relative risk from two to four times for coronary disease, stroke, heart failure, peripheral arterial disease, renal insufficiency, atrial fibrillation and dementia/cognitive impairment. Poorly controlled hypertensive patients have an increased risk for cardiovascular complications 1 .
