Bidirectional Interactions of NK Cells and Dendritic Cells in Immunotherapy: Current and Future Perspective

Abstract: NK cells and dendritic cells (DC) are innate cellular components that regulate adaptive immune responses in the immune surveillance of cancer and infections. Interactions of NK and DC are bidirectional. In this mini review, we summarized how NK cells regulate immature DC editing and maturation, how DC regulate NK-cell functions reciprocally in the NK-DC crosstalk, and the importance of NK-DC crosstalk in antitumor immunity. Enhancing NK-DC crosstalk by cellular factor(s), antibodies or creating a microenvironm… Show more

“…39,43 Moreover cytokines, cellular factors, antibodies promote NK cell activations, and DC-NK crosstalk establishing a microenvironment which enables antitumor reactions. 44,45 Moreover, a crosstalk between CIK cells and DC has been reported recently, demonstrating an influence of a DC stimulation on the phenotype as well as antileukemic cytotoxicity of CIK cells. 46 Recently a crosstalk between DC and iNKT cells has been shown (mediated, eg, by chemokine receptor-expression), leading to an amplification of antitumor-immune reactions.…”

“…Therefore, NK cells may limit or exacerbate immune response; as a result, both cellular interactions and the local environment in which the NK cell resides in may influence its cytotoxic functions. The intensity and the feature of NK cell cytotoxic and cytokines release also depend on the cytokine milieu, as well as on interactions with other cells of the immune system such as macrophages, DCs, and T cells [195,196].…”

“…16,17 DCs are the most efficient APCs that can activate both T cells and B cells and thus, they act as a bridge between innate and adaptive immunity. [18][19][20][21] Besides, DCs also have the ability to inuence T cell polarization via three ways: (i) antigen presentation, (ii) co-stimulatory molecule expression, and (iii) direct contribution by DCs to the immediate cytokine milieu that directs the resultant Th cell response.…”
