Binding and Inhibitory Effect of the Dyes Amaranth and Tartrazine on Amyloid Fibrillation in Lysozyme

Abstract: Interaction of two food colorant dyes, amaranth and tartrazine, with lysozyme was studied employing multiple biophysical techniques. The dyes exhibited hypochromic changes in the presence of lysozyme. The intrinsic fluorescence of lysozyme was quenched by both dyes; amaranth was a more efficient quencher than tartrazine. The equilibrium constant of amaranth was higher than that of tartarzine. From FRET analysis, the binding distances for amaranth and tartrazine were calculated to be 4.51 and 3.93 nm, respectiv… Show more

“…Among food additives approved for human use, azo compounds are negatively charged at physiological pH and are widely used as food colorants in a large variety of foodstuff and pharmaceuticals (Saleh et al ., 2016). They are characterized by aromatic rings in conjunction with N = N linkages and several are known to interact with biomacromolecules (Basu and Kumar, 2015; Basu and Suresh Kumar, 2017; Al-Shabib et al ., 2018). Primarily, six dyes were selected for the combined evaluation of the antibacterial activity.…”

“…Concerning the beneficial effect of Tartrazine, it had an important inhibitory role in fibrillogenesis and showed the potential anti-amyloidogenic property of food colorants (Basu and Kumar 2017).…”

“…The latter include: surfactants, copper/zinc ion chelators [13], flavonoids (e.g., oleuropein aglycone, epigallocatechin gallat, curcumin, etc.) [14][15][16], and organic dyes such as Congo red (CR) [17], azo dyes [18], etc. Among the above compounds, peptide inhibitors seem to be the most promising agents for prevention of fibrillization process in vivo due to their high biocompatibility and affinity for the amyloidogenic core sequence of the fibril-forming proteins [9,19].…”
