Bioactive Polymeric Metallosomes Self-Assembled through Block Copolymer–Metal Complexation

Abstract: Spontaneous formation of polymeric metallosomes with uniform size (~100 nm) was found to occur in aqueous medium through the reaction of an anticancer agent, (1,2-diaminocyclohexane)platinum(II) (DACHPt), with a Y-shaped block copolymer of ω-cholesteroyl-poly(L-glutamic acid) and two-armed poly(ethylene glycol) (PEGasus-PLGA-Chole). Circular dichroism spectrum measurements revealed that the PLGA segment forms an α-helix structure within the metallosomes, suggesting that secondary-structure formation of metallo… Show more

“…D rug-loaded polymeric nanocarriers are of immense interest as anticancer drug-delivery systems due to their ability to extravasate within the tumours via the enhanced permeation and retention (EPR) effect [1][2][3][4] . Polymeric delivery systems can also enhance the aqueous solubility and modify the pharmacokinetics and biodistribution of lipophilic drugs [5][6][7][8][9][10][11][12] .…”

“…Accordingly, such materials as quantum dots12, gold nanoparticles345, carbon nanotubes67, graphene89, and polymersomes10 have all been investigated and have shown promise in a wide range of biomedical applications11. Simultaneous diagnostics, therapeutics, and monitoring of response to treatment are realized with such multifunctional nanomaterials and nanostructures, which also offer the potential of both reducing common chemotherapy- or radiation-associated side effects and increasing the effectiveness of therapy121314.…”

“…Cisplatin, a cytotoxic drug with wide anticancer spectrum was then loaded on GNR-PGA via Please do not adjust margins Please do not adjust margins formation of coordination bonds between the carboxylic groups of PGA and the platinum atom of cisplatin (Scheme 1b). 26,27 A cisplatin to Au loading ratio of 4.8 wt% and a drug loading efficiency of 13.0% were achieved when GNR@Pt nanoparticles were prepared at a cisplatin to Au feeding ratio of 0.37 as determined using inductively coupled plasma mass spectrometry (ICP-MS) measurement ( Figure 1c). The LSPR absorbance of GNR-PGA decreased slightly upon cisplatin loading, which might be caused by sample loss during centrifugation ( Figure 1a & SI 8).…”
