Biochemical parameters to assess cell differentiation of Bouvardia ternifolia Schlecht callus

Abstract: Calli derived from leaves and radicles of B. ternifolia were grown on Murashige and Skoog (MS) basal medium, and the effects of different nitrogen sources on the rate of callus growth and on the enzymes related to nitrogen assimilation were studied. Ammonium alone did not support callus growth unless a Krebs-cycle intermediate was added to the medium. The activities of glutamine synthetase (EC 6.3.1.2), glutamate synthase (EC 1.4.7.1), and glutamate dehydrogenase (EC 1.4.1.2) were measured in homogenates of ca… Show more

“…Similarities in the incorporation of 14C compounds as well as in the profile of sugars and amino acids in tomato callus, leaf and fruit explant have been shown (Danek-Jezik and Schmidt 1979;Obato-Sasamoto et a1 1984). Differences in the biochemical properties in the callus cultures and their explants were pointed out in tobacco and potato tissues with respect to the metabolism of 14C glucose and 14C0, (Dejimenez et al 1983;Abou-Mandour and Kaiser 1984).…”

“…Las hojas y flores (explantes) se pusieron en una disolución 1:1 de ácido ascórbico y cítrico (100 mg/L). Las hojas se cortaron en segmentos de 1 cm 2 y las flores se cortaron en cuatro segmentos colocándolas en el medio de cultivo Murashige and Skoog (MS) (Sánchez & Fernández, 1983)…”
