Biodegradable Charge‐Transfer Complexes for Glutathione Depletion Induced Ferroptosis and NIR‐II Photoacoustic Imaging Guided Cancer Photothermal Therapy

Abstract: Suffering from the laborious synthesis and undesirable tumor microenvironment response,t he exploitation of novel NIR-II absorbing organic photothermal agents is of importance to promote phototherapeutic efficacy.Herein, two kinds of charge-transfer complex nanoparticles (TMB-F4TCNQ and TMB-TCNQ) are prepared by supramolecular assembly.Because of the larger energy gap between donor and acceptor,T MB-F4TCNQ presents higher charge-transfer degree (72 %) than that of TMB-TCNQ (48 %) in nanoaggregates.T herefore,T… Show more

“…H 2 O 2 ) and antioxidants (e.g., GSH) directly influence the survival state of tumor cells, the GSH‐induced drug release behavior reminded us FIT NPs might born to effective depletion of GSH in tumor cells and broke the redox equilibrium, which was further considered as an adjuvant therapy for accelerating the apoptosis of tumor cells. [ 14 ] To verify this point, a ThiolTracker Violet reagent was used to trace the change of intracellular GSH levels in CT26 tumor cells (Figure 1H and Figure S5, Supporting Information). We incubated FIT NPs with cells under a time increasing sequence, the quantitative FL intensity of ThiolTracker Violet of control group was 3.9‐fold higher than that of the 6 h incubation group, indicating FIT NPs could significantly reduce the content of GSH in tumor cells.…”

“…Dysregulated ferroptosis has been evidenced to be implicated in the pathogenesis and progression of many human diseases [ 196 ]. Furthermore, targeting induction of ferroptosis provides a potential therapeutic strategy for the clinical intervention of cancers, especially the other traditional therapy-resistant cancers [ 197 , 198 ]. As mentioned above, the major features of ferroptosis have been extensively observed in the diseased mucosa in patients and animal models.…”

“…Since the ferroptotic process is rapid and intense without multi-level signal transduction unlike apoptosis and immune responses, it has garnered considerable attention ( Fei et al, 2020b ). When ferroptosis-inducing strategies and ferroptosis-based nanotherapeutics were integrated for HCC therapy, remarkable tumor suppression was achieved during the last decade ( Ou et al, 2021 ). In all the attempts to explore ferroptotic treatment, metal elements such as iron, Mn, and copper, etc., have been found to be superior in inducing tumor cell ferroptosis, promoting ROS production, and interfering with cell communication when acting as Fenton or Fenton-like biocatalysts ( Huo et al, 2019 ; Lin et al, 2019 ).…”
