Biodegradable Polymeric Architectures via Reversible Deactivation Radical Polymerizations

Abstract: Reversible deactivation radical polymerizations (RDRPs) have proven to be the convenient tools for the preparation of polymeric architectures and nanostructured materials. When biodegradability is conferred to these materials, many biomedical applications can be envisioned. In this review, we discuss the synthesis and applications of biodegradable polymeric architectures using different RDRPs. These biodegradable polymeric structures can be designed as well-defined star-shaped, cross-linked or hyperbranched vi… Show more

“…Ring opening polymerization (ROP) provides polymers that are biocompatible as well as biodegradable such as poly lactides, polycaprolactones, polycaprolactams, poly esteramides, polyamides etc. [1][2][3][4] While most of these poly merizations require metal catalysts and rather harsh conditions, the poly merization of N-carboxyanhydrides (NCA) is a versatile method which either proceeds through metal complex initiators or by primary amines. Metal complexes are removed from the polymers after of S-(o-nitrobenzyl)-l-cysteine polymer lead to the formation of stable nanoparticles.…”

“…Mammals’ kidney filtration is limited by the molar mass and shape of a water‐soluble polymer, with the rough cutoff at M n ≈ 30 000, resulting in much longer circulation time in blood above that threshold. [ 58 ] Pharmaceuticals based on PEGylated species (PEG: poly(ethylene glycol)) are very common and approved by the US Food and Drug Administration (FDA). However, prolonged contact with PEGylated species can lead to the formation of anti‐PEG antibodies, with reported negative health outcomes.…”

“…207 To confer biodegradability to polymers, they must be designed with intra-linkers that can be cleaved by either enzymatic catalysis or physiological substances (e.g., glutathione). 208 Biodegradable linkages can be tailored on the polymer backbones, on the side chains, on the cross-linking agents, etc. RROP of CKAs is used to incorporate ester functionalities (e.g., caprolactone units) into the vinyl polymer backbone enabling degradation by hydrolysis of the polymer backbone, which is impossible for vinyl polymers with only C-C bonds in the backbone (Fig.…”
