BioPhotonics workstation: A versatile setup for simultaneous optical manipulation, heat stress, and intracellular pH measurements of a live yeast cell

Abstract: In this study we have modified the BioPhotonics workstation (BWS), which allows for using long working distance objective for optical trapping, to include traditional epi-fluorescence microscopy, using the trapping objectives. We have also added temperature regulation of sample stage, allowing for fast temperature variations while trapping. Using this modified BWS setup, we investigated the internal pH (pHi) response and membrane integrity of an optically trapped Saccharomyces cerevisiae cell at 5 mW subject t… Show more

“…3.8(b)). This allows room for a variety of auxiliary applications such as advanced micro-spectroscopic [26] or multi-photon characterization methods or pH mapping of heat stressed cells through fluorescence ratio imaging [70]. This has also allowed direct side view imaging, making three dimensional experiments more intuitive ( Fig.…”

“…We tested the idea of optically steerable freestanding waveguides using our BioPhotonics Workstation (BWS). 4 BWS uses real-time reconfigurable counter-propagating beam traps controlled by direct spatial mapping from an addressable light-shaping module. Axial manipulation is achieved by balancing the intensity ratios of the counterpropagating beams.…”
