Biosensor-based comparison of the ecotoxicological contamination of the wastewaters of Southern Russia and Southern Germany

Abstract: To assess the ecotoxicological and sanitary situation in two European metropolis, Rostov-on-Don (Southern Russia) and Munich (Southern Germany), wastewaters of the two cities were examined with a panel of bacterial lux-biosensors:The presence of different genotoxic compounds and substances with the oxidative and membrane-damaging effects was revealed in contaminated wastewater with the applied panel of the lux-biosensors. The integral toxicity was approximately the same in both cities but demonstrated opposite… Show more

“…The oxyR gene promoter is induced by intracellular hydrogen peroxide; the addition of 4a to the medium leads to the appearance of peroxide inside E. coli cells. This can occur both during the oxidation of 4a with atmospheric oxygen (the effect is typical for some hydrocarbons [23]), resulting in the formation of reactive oxygen species in the environment, or when a compound enters the cell and interacts with various cellular components, such as proteins of the respiratory chain. Compounds do not induce DNA damage that stops the replication fork (SOS response), do not cause the transcriptional activation of DNA glycosylase AlkA (no alkylation), do not cause oxidative damage by superoxide anion radical (i.e., do not activate P soxS ) and do not cause the activation of ferritin gene promoter, which protects DNA from oxidative damage.…”

“…Nowadays, biosensors are rapidly developing toward higher sensitivity, specificity, reproducibility, automation and high throughput (Conforti et al, 2008 ; Skotti et al, 2014 ; Yang et al, 2016 ). Furthermore, some specific biosensors with recombinant bioluminescent bacteria have also been designed to detect cell damage, like DNA damage, protein damage, cell membrane damage and stress response (Ahn et al, 2009 ; Sazykin et al, 2016 ; Jiang et al, 2017 ).…”
