This experiment aimed to investigate the effect of dietary Nigella sativa on cell mediated immune response. A total of eighteen male Wistar rats were divided equally into control and black seeds at 30 and 50 g/kg/diet (Sa30 and Sa50), respectively for 30 days. Weight gain, feed intake, feed conversion ratio (FCR), cell mediated immune response was monitored after injection of 0.1 mL of 10% phytohemagglutinin (PHA). Intumesce Index, serum total antioxidant capacity (TAC), Catalase (CAT), Interleukin-12 (IL-12), gamma interferon (γ-IF) and tumor necrosis factor alpha (TNF-α) were determined. Histopathological examination and immunohistochemistry of splenic caspase-3 and CD 8 were done. Nigella sativa significantly (p<0.05) improved weight gain and FCR in Sa30 and Sa50 groups. Significant increase for Intumesce Index in Sa50 group was observed. Total TAC, CAT, IL-12, γ-IF and TNF-αincreased significantly (P<0.05) in Sa30 and Sa50 groups. Histological examination of PHA stimulated foot pads showed more leukocytes infiltration and edema in a dose dependent pattern. Splenic caspase-3 and CD 8 showed significant (P<0.05) decrease and increase, respectively in Sa30 and Sa50 groups. Nigella sativa seeds had immunostimulatory ability through their antioxidant potential, cytokines induction, CD 8 promotion and reducing splenic apoptosis.

Background: Enhancing the immune system is very important during the Covid-19 pandemic to prevent infections and reduce the risk of disease severity. Therefore, it is necessary to use natural products with an immunostimulant effect. In Islam, there is a system of treatment or disease prevention based on Al-Quran and Hadith, which is called Tibb an-Nabawi or prophetic medicine. Objective: This research aims to conduct a literature study of Tibb an-Nabawi, which has proved to be able to increase the immune system, based on Islamic and scientific approaches. Method: The research was based on a systematic literature review using research articles from the last ten years. The inclusion criteria were articles discussing the immunostimulatory activity of Tibb an-Nabawi, while the exclusion criteria were immune system enhancing herbs that were not Tibb an-Nabawi. Results: Based on the literature studies, it is known that at least six natural products based on Tibb an-Nabawi have scientifically proved to increase the immune system, namely black cumin, honey, dates, ginger, garlic, and pumpkin. The mechanism of immune system enhancement is different in each substance, but in general, they can increase humoral or cellular immunity. The active compounds contained in each of these ingredients contribute to the resulting immunostimulant activity. Conclusion: Black cumin, honey, dates, ginger, garlic, and pumpkin are natural products based on Al-Quran and Hadith, which have scientifically proved to enhance the immune system. Oxidative stress in metabolic syndrome (MS) results in vascular endothelial damage, generates inflammatory reactions, and increases the risk of impaired kidney function. Black cumin seed oil (BCSO) is thought to be able to improve the immune response and protect damage to the kidneys. The purpose of this study was to determine the effect of BCSO on TNF-α and creatinine levels in patients at risk of metabolic syndrome with standard therapy at the Jetis I Public health center (PHC). This study uses stored biological material (BM) of patients at risk of outpatient metabolic syndrome in Jetis I PHC, Bantul, for the period July 2017 to July 2018. Researchers used 61 blood specimens of patients at risk of MS, 30 of patients at risk of MS with a combination of standard therapy +BCSO, and 31 patients at risk of MS with standard + placebo combination therapy. The administration of BCSO has been done for 20 days. On the 21st day, a blood sample was taken. Blood creatinine levels were measured by the Jaffe method. TNF-α levels were determined by the enzyme-linked immunosorbent assay (ELISA) method in the Clinical Pathology Laboratory of the Faculty of Medicine UGM. Differences in TNF-α levels and creatinine levels between groups were analyzed using independent samples ttest (95% confidence level). TNF-α levels did not differ between the two groups (p> 0.05). The TNF-α levels of the BCSO group were 101.43 ± 22.89 nmol/ml, and the placebo group 106.73 ± 30.03 nmol/ml. There was no statistically significant difference in creatinine levels between the two groups (p <0.05), but it was clinically significant. The creatinine content of the BCSO group was 1.16 ± 0.19 mg/ml, and the placebo group was 1.35 ± 0.32 mg/ml with a p value> 0.05, but it was clinically significant. Based on the results of this study, it can be concluded that the administration of BCSO additional therapy has no effect on TNF-α levels but has an impact on creatinine levels in patients at risk of metabolic syndrome in Jetis I primary care center, Bantul Yogyakarta.
