BLIMP-1: trigger for differentiation of myeloid lineage

Abstract: B lymphocyte-induced maturation protein-1 (BLIMP-1 or PRDI-BF1) is induced when bone marrow-derived progenitors differentiate in response to macrophage-colony stimulating factor (M-CSF) and is present in peripheral blood monocytes and granulocytes. BLIMP-1 is also induced during differentiation of U937 and HL-60 cells into macrophages or granulocytes. Induction of BLIMP-1 mRNA during macrophage differentiation of U937 and HL-60 shows a biphasic pattern. Overexpression of BLIMP-1 is sufficient to initiate macro… Show more

“…PRDM1/Blimp-1 acts as a transcriptional repressor through competitive promoter occupancy (13,19), or by recruiting histone deacetylase activity (20,21) and the histone H3 lysine-9 methyltransferase, G9a (22). In mature tissues, PRDM1/Blimp-1 is additionally expressed in squamous epithelia (23), macrophages (24), and T cells (25)(26)(27). However, targets of PRDM1/Blimp-1 specifically relevant to these lineages are currently undefined.…”

“…Prdm1-induced maturation of lymphocytes is correlated with the ability of Prdm1 to repress numbers of genes critical for mature B cell identity and cellular proliferation [5,[8][9][10][11][12][13]. Prdm1 is also needed for the activation of T cells [14][15][16][17][18], the differentiation of myeloid lineage [19] and the maturation of nature killer (NK) cells [20].…”
