Blood Sampling in the Rat: Current Practices and Limitations

Abstract: Historically, blood samples have been taken by puncture of the orbital sinus under light ether anaesthesia. In the last decade, there was concern over the use of ether as an anaesthetic. Furthermore, in recent years, more attention has been focused on the need to improve current laboratory practices for the bene¢t of animal welfare. Exposure for 2 min to a CO 2 /O 2 mixture produced acute hyperkalaemia and changes in a range of haematological parameters. Hyperkalaemia in some individuals reached pathological l… Show more

“…The prolonged bleeding and blood sample collection also might lead to mild anemia which is reflected in the CBC results (Tables S5–S7). Most of the anemia symptoms (the reduction of platelet count, polychromasia, and anisocytosis) were observed before and after the investigation in male and female rats. Although the reduction of HGB and HCT was found after the study, the parameters were already near the baseline before the multiple-dose study (Table S5).…”

“…Anaesthesia, specifically isoflurane, can have an effect on the percentage of neutrophils found in C3H mice, with 30 min exposures leading to a 15.4 % reduction in the number of circulating WBCs, and specifically a 26.9 % reduction in neutrophils up to 48 h after exposure (Colucci et al 2013 ; Jacobsen et al 2004 ). Exposure to 4 % isoflurane, if administrated for a duration longer than 5 min may also have had an effect on erythrocytes parameters (Nahas and Provost 2002 ). The length of time the murids in this study were under anaesthesia is unknown.…”
