BMP3 Affects Cortical and Trabecular Long Bone Development in Mice

Abstract: Bone morphogenetic proteins (BMPs) have a major role in tissue development. BMP3 is synthesized in osteocytes and mature osteoblasts and has an antagonistic effect on other BMPs in bone tissue. The main aim of this study was to fully characterize cortical bone and trabecular bone of long bones in both male and female Bmp3−/− mice. To investigate the effect of Bmp3 from birth to maturity, we compared Bmp3−/− mice with wild-type littermates at the following stages of postnatal development: 1 day (P0), 2 weeks (P… Show more

“…Furthermore, BMP3 serves as a target of miR-450b to regulate bone formation [ 37 ]. A recent study has presented that bone mineralization was enhanced in BMP3−/− mice, suggesting that BMP3 could influence long bone development in mice [ 38 ]. In this study, experiments of the luciferase assay showed that the suppression of miR-181a-5p increased luciferase activity while the overexpression decreased it.…”

“…The scanning resolution was set at 18 μm and 3D reconstruction of trabecular bone was used by Ctvol software. Parameters computed from these data include trabecular thickness (Tb.Th), trabecular number (Tb.N), trabecular separation (Tb.Sp), trabecular pattern factor (Tb.Pf), bone volume fraction (BV/TV) and bone surface density (BS/TV) [ 38 , 42 ].…”

“…In vitro experiments using pluripotent mesenchymal progenitor C3H10T1/2 cells and preosteoblastic C2C12 cells showed that BMP-2, -6, and -9 exhibit high ability to induce both early and late osteogenic markers as well as matrix mineralization, while most BMPs can effectively promote the terminal differentiation of committed osteoblastic precursors and osteoblasts ( 18 , 19 ). In contrast to other BMPs, BMP3 has been proposed to act as an inhibitor of osteogenic BMPs and antagonizes the osteogenic activity of BMP-2, -4, -6, -7, and -9 ( 20 ), while deletion of Bmp 3 gene results in increased skeletal bone volume ( 21 ).…”

“…BMP2 is a primary participant in postnatal skeletal homeostasis through its regulation of SSC niches and BMP2 signaling is required for the di erentiation of osteoprogenitor cells into the fully functioning osteoblasts (50). On the other hand, the role of BMP3 has never been fully elucidated (51)(52)(53) but recent discoveries presented at the Conference highlighted a novel role of BMP3 in enabling chondrocyte di erentiation following decreased expression of BMP3 and ALK2.…”
