Bone morphogenetic protein-7 represses hepatic stellate cell activation and liver fibrosis via regulation of TGF-β/Smad signaling pathway

Abstract: BACKGROUND Liver fibrosis is a refractory disease whose persistence can eventually induce cirrhosis or even liver cancer. Early liver fibrosis is reversible by intervention. As a member of the transforming growth factor-beta (TGF-β) superfamily, bone morphogenetic protein 7 (BMP7) has anti-liver fibrosis functions. However, little is known about BMP7 expression changes and its potential regulatory mechanism as well as the relationship between BMP7 and TGF-β during liver fibrosis. In addition, the … Show more

“…The high ratio of liver/body weight induced by CCl 4 was reduced by treatment with celastrol (Figure C). As continuous administration of CCl 4 could lead to liver fibrosis, therefore, we then studied the effect of celastrol on CCl 4 ‐induced liver fibrosis in rats. Celastrol decreased the collagen deposition caused by CCl 4 injection (Figure A).…”

“…Bone morphogenetic protein 7 (BMP-7) is a member of the transforming growth factor-beta (TGF-β) superfamily [ 65 ]. Clinically, BMP-7 is mostly used in the research and application of plastic surgery, such as that treating long bone nonunion fractures [ 66 ].…”

“…The typical characteristics of hepatic fibrosis are the activation of HSC and excessive deposition of ECM 3 . As a unique marker of HSC activation, a-SMA is positively correlated with proliferation of HSCs and degree of liver fibrosis [28]. Accordingly, we detected expression of a-SMA by immunohistochemistry.…”
