Bone Reconstruction Using Two-Layer Porcine-Derived Bone Scaffold Composed of Cortical and Cancellous Bones in a Rabbit Calvarial Defect Model

Abstract: In this study, we aimed to investigate the bone regeneration efficiency of two-layer porcine-derived bone scaffolds composed of cancellous and cortical bones in a rabbit calvarial defect model. Four circular calvaria defects were formed on cranium of rabbit and were filled with block bone scaffolds of each group: cortical bone block (Cortical group), cancellous bone block (Cancellous group), and two-layer bone block (2layer group). After 8 weeks, new bones were primarily observed in cancellous parts of the Can… Show more

“…Thus, non-synonymous SNPs in mtDNA may yield a substantial quantity of mutant peptides, offering an additional source of neoantigens. [49][50][51] INDELs INDEL mutations are mainly caused by the insertion or deletion of base pairs in the genome, which frequently lead to nonsynonymous novel ORFs, also known as frameshift mutations. 27,52 Frameshift INDELs can generate more types of neoantigens with increased MHC-I binding affinity, suggesting a higher immunogenic mutation type compared to SNVs (Table 2).…”

“…From the natural bone structure, this study designs porous scaffolds with a biomimetic design perspective by using different inner and outer pore units. The outer pore units adopt BCC structure to simulate the weight‐bearing function of human cortical bone, while inner pore units using TPMS structure to obtain a good permeability and simulates the inner layer of cancellous bone 30,31 . The main advantage of the biomimetic porous structure proposed in this study is that it can mimic the cortical and trabecular bone structures of natural bone tissue, which is beneficial for adjusting mechanical properties while maintaining a high permeability for transportation of nutrients and metabolic waste.…”

“…Before proceeding with the descriptions of the analyses, it is important to justify the non-performance of the group with defects filled by blood clot (BC) in this study, as we prioritize the principle of the three Rs, as proposed by reduction, substitution, and refinement in the use of animals, with the objective of constantly promoting a balance between scientific progress and animal welfare [ 45 , 46 , 47 ]. Likewise, because such a group has already been carried out in our previous studies with the same methodology, animal model, and experimental period (volume density of the newly formed bone, mean 7.06 ± 0.49 and bone volume, mean 5.20 ± 1.02) [ 21 ].…”
