Brain‐derived neurotrophic factor ameliorates lipid metabolism in diabetic mice

Abstract: Taken together with the accelerating effect of BDNF on energy metabolism, these findings indicate that BDNF improves glucose and lipid metabolism in obese diabetic animals without enlarging liver or adipose tissues.

“…BDNF increases fat oxidation in an AMPK-dependent manner in L6 myotubes Having established that BDNF was a contraction-induced protein, we next sought to determine whether BDNF affected signalling pathways associated with fat oxidation in skeletal muscle. This hypothesis was based on the observation that BDNF reduces obesity in diabetic mice [18]. AMPK phosphorylates ACCβ resulting in inhibition of ACC activity, which in turn leads to a decrease in malonyl-CoA content, relieving inhibition of carnitine palmitoyl transferase 1 and increasing fatty acid oxidation (FAO).…”

“…It is also known that BDNF reduces food intake and lowers blood glucose in genetically modified (db/db) obese mice [15][16][17][18]. It is clear, therefore, that BDNF plays a role in both neurobiology and metabolism.…”

“…In addition to its neurotrophic and synaptotrophic actions, including the promotion of growth and survival in neurons, 7,8 BDNF plays a role in learning and memory, 9 the regulation of food intake, 10 glucose and lipid metabolism and energy homeostasis. 11,12 BDNF is present in the nervous system and peripheral tissues and is also found in blood. [13][14][15] Accumulating evidence shows the serum BDNF levels to be associated with psychiatric and metabolic disorders, including depression, 16,17 Alzheimer disease, 18 and diabetes mellitus.…”

“…Circulating BDNF was reported to correlate positively to LDL cholesterol, total cholesterol and triglyceride levels in humans (16,17). In animal experiments, BDNF treatment reported to lower blood glucose, non-esterified free fatty acid, total cholesterol and phospholipid levels significantly in diabetic models, along with liver weight, liver triglyceride contents and fatty liver in histological examinations (28). Furthermore, administration of simvastatin elevated the expression of BDNF in the brain and accelerated the neuronal and functional recovery (29).…”
