BRD7 Acts as a Tumor Suppressor Gene in Lung Adenocarcinoma

Abstract: Lung cancer is one of the most malignant tumors and the leading cause of cancer-related deaths worldwide. Among lung cancers, 40% are diagnosed as adenocarcinoma. Bromodomain containing 7 (BRD7) is a member of bromodomain-containing protein family. It was proved to be downregulated in various cancers. However, the role of BRD7 in lung adenocarcinoma is still unknown. Western blot and qRT-PCR was performed to measure the BRD7 expression in lung adenocarcinoma tissues and cells. CCK8 and migration assay was done… Show more

“…BRD7 is a multifunctional gene, exhibiting its functions through several pathways (6). As a tumor suppressor, BRD7 inhibited cancer cell growth via phosphoinositide 3-kinase (PI3K)/Akt, ER stress, ERK and hypoxia-inducible factor α (HIF1α)/lactate dehydrogenase A signaling (10,3436). Furthermore, BRD7 regulated glucose homeostasis through glycogen synthase kinase 3β and X-box binding protein 1 (37,38).…”

“…These data may indicate that BRD7 is associated with PCa progression. In addition to PCa, upregulation of BRD7 has been associated with decreased cell proliferation and migration in lung adenocarcinoma and may exert antitumor effects by activating the phosphorylation of ERK (34). The current study provides evidence that BRD7 may be used as an efficient biomarker for PCa diagnosis and prognosis.…”

“…Lung cancer, with high morbidity and mortality rate, ranks first among all the malignancies in developed and developing countries such as India, Europe, China and the USA [ 41 ]. Various tumor suppressor genes associated with lung cancer include, tumor suppressor gene TP53 [ 42 ], Adenomatous polyposis coli (APC) [ 43 ], Bromodomain containing 7 (BRD7) [ 44 ], Cyclin Dependent Kinase Inhibitor 2A (CDKN2A) [ 45 ] and NM23 [ 46 ]. Proto-oncogenes constituting lung cancer are receptor tyrosine kinase (RTK) [ 47 ], MYC [ 48 ], JUN [ 49 ], FOS [ 50 ], RAF1 [ 51 ] and ERBB1&2 [ 52 ].…”
