C-kit receptor in the human vas deferens: distinction of mast cells, interstitial cells and interepithelial cells

Abstract: The molecular mechanisms underlying the regulation of vas deferens (VD) motility and semen emission are still poorly understood. Interstitial cells of Cajal (ICC), which harbour the c-kit receptor (CD117), provide the basis of coordinated gut motility. We investigated whether c-kit receptor-positive cells also exist in the normal human VD. Enzyme and fluorescence immunohistochemical techniques were applied on serial sections of human proximal, middle, and distal VD segments (nZ49) employing 13 different monocl… Show more

“…Finally, our data confirm and expand the set of negative excretory duct system IHC markers to include PSA, PSMA, WT1, calretinin, OCT4, SALL4, and CD117. 5,17,18,21,22 TV is a mesothelial-derived cell layer that forms the outer surface of the testis and lines the potential space of the scrotal sac. 2 Our data indicate that TV expresses CK7, WT1, and calretinin, which is concordant with previously published data and supports its mesothelial origin.…”

“…We initially used c‐kit immunohistochemistry similar to previous studies 9,10,12 but we subsequently adopted a double immunofluorescence technique to better distinguish mast cells from ICC. Problems with autofluorescence were overcome by reducing primary antibody concentrations and incubation periods and the preferential selection of surgical specimens; the use of Sudan black B was unhelpful 22 . No obvious differences between monoclonal and polyclonal c‐kit antibody immunolabeling emerged.…”

“…8 C-kit immune positive-stained cells have also been demonstrated in the urinary bladder, urethra, prostatic stroma, vas deferens, corpus cavernosum and spermatogonia. [10][11][12][13][14][15] We investigate the quantitative and ultrastructural changes of ICC UPJ obstruction using light and electron microscopy.…”
