Calcium homeostasis in the outer segments of retinal rods from the tiger salamander.

Abstract: SUMMARY1. The processes regulating intracellular calcium in the outer segments of salamander rods have been investigated. The main preparation used was the isolated rod loaded with the Ca2+-sensitive photoprotein aequorin, from which outer segment membrane current and free [Ca2+]i could be recorded simultaneously. Two other preparations were also used: outer segment membrane current was recorded from intact, isolated rods using a suction pipette, and from detached outer segments using a whole-cell pipette. 11.… Show more

“…Effect of a continuous change of temperature on the light-sensitive current for six cells in which a similar protocol to that described in Fig. 1 (Lagnado et al 1992). This effect will be larger at higher temperatures where the light-sensitive current is greater.…”

“…In salamander rods the time constant of relaxation of the exchange current is 700 ms (Lagnado et al 1992) and the outer segment diameter of 11 ,um is 7 times larger than the value of 1P5 /tm in rats, where the time constant is about 100 ms (see p. 469). It appears therefore that the different time constants in different species may be largely accounted for by differences in outer segment diameter.…”

“…Decreases in cytosolic calcium concentration in ROS also occur as a result of photoreceptor stimulation (Yau and Nakatani, 1985;McNaughton, Cervetto, and Nunn, 1986;Ratto, Payne, Owen, and Tsien, 1988;Lagnado, Cervetto, and McNaughton, 1992;Younger, McCarthy and Owen, 1992). This decrease is attributed to closure of the cGMP-gated ion channel preventing calcium entry into the outer segment, while the plasma membrane Na-Ca-K exchanger continues to extrude intracellular calcium (for review, see McNaughton, 1990).…”
