Candidate gene association study conditioning on individual ancestry in patients with type 2 diabetes and metabolic syndrome from Mexico City

Abstract: Background Type 2 diabetes (T2D) is influenced by diverse environmental and genetic risk factors. Metabolic syndrome (MS) increases the risk of cardiovascular disease and diabetes. We analysed 14 cases of polymorphisms located in 10 candidate loci, in a sample of patients with T2D and controls from Mexico City.

“…2016;60/3 TCF7L2 (transcription factor 7-like 2) is among the highly replicated genes showing robust association with T2DM (8,9). The TCF7L2 gene spans 215.9 kb on chromosome 10q25.3, and its product is a high-mobility box-containing transcription factor involved in the Wnt signaling pathway (10).…”

“…The subsequent genome-wide association analysis on T2DM showed a strong signal for TCF7L2 in the French population [23]. This prompted inclusion of TCF7L2 gene variants in a large number of association studies in various populations world-wide [24][25][26][27][28][29]. Furthermore, a number of meta-analyses supported this robust finding [30,31].…”

“…However, inconsistent results on the association between the above variant alleles and T2DM risk may likely be due to the studies being carried out in individuals of different ethnicity [24,26]. The ADRB3 rs4994 polymorphism however, is reported to be associated with T2DM from Mexico city [27]. Moreover, Several past studies have connected this non-synonymous rs4994 polymorphism to T2DM, obesity, insulin resistance and hypertension [28][29][30][31].…”
