CZE is a well‐established technique for charge heterogeneity testing of biopharmaceuticals. It is based on the differences between the ratios of net charge and hydrodynamic radius. In an extensive intercompany study, it was recently shown that CZE is very robust and can be easily implemented in labs that did not perform it before. However, individual characteristics of some examined proteins resulted in suboptimal resolution. Therefore, enhanced method development principles were applied here to investigate possibilities for further method optimization. For this purpose, a high number of different method parameters was evaluated with the aim to improve CZE separation. For the relevant parameters, design of experiments (DoE) models were generated and optimized in several ways for different sets of responses like resolution, peak width and number of peaks. In spite of product specific DoE optimization it was found that the resulting combination of optimized parameters did result in significant improvement of separation for 13 out of 16 different antibodies and other molecule formats. These results clearly demonstrate generic applicability of the optimized CZE method. Adaptation to individual molecular properties may sometimes still be required in order to achieve optimal separation but the set screws discussed in this study [mainly pH, identity of the polymer additive (HPC versus HPMC) and the concentrations of additives like acetonitrile, butanolamine and TETA] are expected to significantly reduce the effort for specific optimization.

Capillary electrophoresis (CE) continues its invasion into the domain of inorganic analysis. This review summarizes the progress and significant developments of CE of inorganic ions over the first two years of the new century. As manifested by the substantial number of published articles, this field is still witnessing a growing activity of many separation scientists. To provide an in-depth treatment of fundamental and methodological aspects of inorganic ion analysis by CE, an extensive fraction of the relevant literature is covered whereas only most innovative application reports are briefly considered. Most of the recent advancements have focused on CE system modernization designed to improve a limited detection sensitivity, which still remains a major obstacle to overcome in order to ensure the method's wider practical acceptance. Attention is also given to how and in which directions the future research could be performed to implement CE in routine inorganic analysis.

Despite the availability of commercial capillary electrophoresis systems for over ten years, where quantitative analysis is required, capillary zone electrophoresis (CZE) has often failed to replace ion chromatography as the method of choice for a large number of analytes, not least inorganic anions. To investigate the reasons for this apparent failing, a review is presented of work that has been carried out to-date involving the quantitative application of CZE to the determination of inorganic anions in industrial and environmental samples. This review summarizes work both investigating and improving the quantitative aspects of the CZE of inorganic anions. A complete survey of how CZE has been applied to the determination of inorganic anions in real samples is given, including what, if any, analytical performance parameters were investigated and quoted, and if quality assurance data and validation methods were briefly considered, thoroughly investigated or simply ignored.
