Cbl-b inhibits P-gp transporter function by preventing its translocation into caveolae in multiple drug-resistant gastric and breast cancers

Abstract: The transport function of P-glycoprotein (P-gp) requires its efficient localization to caveolae, a subset of lipid rafts, and disruption of caveolae suppresses P-gp transport function. However, the regulatory molecules involved in the translocation of P-gp into caveolae remain unknown. In the present study, we showed that c-Src dependent Caveolin-1 phosphorylation promoted the translocation of P-gp into caveolae, resulting in multidrug resistance in adriamycin resistant gastric cancer SGC7901/Adr and breast ca… Show more

“…Mechanistic studies have suggested that redox signals modulate the transporters of membrane proteins 3638 . However, the underlying biochemical process remains largely unclear regarding how ADR perturbs redox homeostasis and the identity of the crucial factor that dominates the increased expression of P-gp.…”

“…Transfections of Cbl-b WT constructs. Transfections of Cbl-b WT constructs were performed as described previously [11,19]. SGC7901/Adr and MCF-7/Adr cells were seeded at 4 × 10 5 cells/well in six-well plates overnight, then the cells were transfected by lipofectamine 2000 reagent with pcDNA 3.1 plasmid including full-length Cbl-b.…”

“…Immunohistochemistry. Immunohistochemical staining for Cbl-b was performed using a streptavidin-peroxidase procedure as described previously [11,19]. Antibody against Cbl-b was from Santa Cruz Biotechnology (USA).…”

“…IHC staining was performed as described previously [18]. An anti-human LBH rabbit antibody was used at a dilution of 1: 100 (ab122223; Abcam, Cambridge, UK); phosphate-buffered saline was used as a negative control.…”

“…Protein was extracted from the cells and quantified as described previously [18, 19]. Equal amounts of protein were separated by electrophoresis on sodium dodecyl sulfate polyacrylamide gel electrophoresis gels, then transferred to polyvinylidene fluoride membranes.…”
