CCL21 promotes the migration and adhesion of highly lymph node metastatic human non-small cell lung cancer Lu-99 in vitro

Abstract: To develop new therapy strategies for lung cancer, we established an animal model, which reflects the clinical features of mediastinal lymph node metastasis of lung cancer. This study was designed to determine whether CCL21 induced biological functions associated with the metastasis of highly lymph node metastatic human non-small cell lung cancer (NSCLC) selected by our model. Orthotopic intrapulmonary implantation of human NSCLC (Lu-99 and A549) was performed to analyze the metastatic characteristics of these… Show more

“…Chemokines and their corresponding receptors have established roles in directing cell migration in many different types of cancer; and expression levels of these proteins have also correlated with clinical outcomes [24][25][26][27][28][29]. The role of CCL25-CCR9 signaling in modulating metastasis was initially reported by Letsch There is an absence of CCL25 staining within the normal architecture of the pancreatic duct and stroma c. In the malignant pancreatic lesion, with characteristic nuclear crowding, enlarged nuclei, and rare mitoses, there is moderate heterogeneous immunostaining for CCL25 protein in the nuclei.…”

“…Signal transduction through these proteins leads to the activation of g proteins and phospholipase c and the elevation of cytosolic free calcium. The chemokine ccL21 induces the adhesion of non-small lung cancer cells to vascular cell adhesion molecule-1 (vcAM-1) by activating integrin through a g protein-coupled receptor (22).…”

“…Migration was assayed in Transwell filter insert chambers (8 µm pore size; corning costar) as previously described (4,11,22). The upper surface of the chamber contained a Transwell filter (8-µm pore size polyvinyl pyrrolidone-free).…”

“…The migration assay was performed in Transwell cell culture chambers (Corning Incorporated Life Sciences, Tewksbury, MA, USA), as reported previously, with some modifications (9). Polyvinylpyrrolidone-free polycarbonate (PVFP) filters (8.0 µm pore size; Nuclepore, Pleasanton, CA, USA) were pre-coated with 1 µg fibronectin (Asahi Technoglass Co., Tokyo, Japan) on the lower compartment.…”
