CD117+ Dendritic and Mast Cells Are Dependent on RasGRP4 to Function as Accessory Cells for Optimal Natural Killer Cell-Mediated Responses to Lipopolysaccharide

Abstract: Ras guanine nucleotide-releasing protein-4 (RasGRP4) is an evolutionarily conserved calcium-regulated, guanine nucleotide exchange factor and diacylglycerol/phorbol ester receptor. While an important intracellular signaling protein for CD117+ mast cells (MCs), its roles in other immune cells is less clear. In this study, we identified a subset of in vivo-differentiated splenic CD117+ dendritic cells (DCs) in wild-type (WT) C57BL/6 mice that unexpectedly contained RasGRP4 mRNA and protein. In regard to the biol… Show more

“…We speculate that hypomethylation over the PARP9 gene body may contribute to lower innate immunity against Mtb and result in greater disease severity. RAS guanyl releasing protein 4 (RASGRP4) is required for the activation of both T cell and neutrophil, and for mast cells and dendritic cells to optimally induce acute natural killer cell-dependent interferon-gamma production in response to LPS [22][23][24]. We speculate that the RASGRP4 gene is up-regulated in early immunological responses independent of its DNA methylation changes, and its hypermethylation over the promoter region at the later stage may contribute to counteracting the early immunological responses.…”

“…We highlighted 9 candidate intolerant genes without a known disease association harboring ultrarare LOF variants in cases but not controls (eTable 21 in the Supplement). CTTN (OMIM 164765) is implicated in lung disease among patients of African descent, RSBN1L (HGNC 24765) is upregulated in COVID-19, and RASGRP4 (OMIM 607320) regulates dendritic and mast cell function …”

“…We chose to further investigate the link between RASGRP4 gene mutations and resistance, because no link to DLBCL had been described. Two closely related GEF, RASGRP1 and RASGRP3 are activated downstream of the B cell receptor, but RASGRP4 is only known to be important for mast cells, certain dendritic cells and neutrophils 29 , 50 , 51 . RASGRP4 is the GEF that mediates GPCR-stimulated RAS activation in unprimed, but not primed, neutrophils 31 , 52 .…”
