CD133 positive progenitor endothelial cell lines from human cord blood

Abstract: Endothelial progenitor cells (EPCs) modulate postnatal vascularization and contribute to vessel regeneration in adults. Stem cells and progenitor cells were found in umbilical cord blood, bone marrow, and mobilized peripheral blood cells, from where they were isolated and cultured. However, the yield of progenitor cells is usually not sufficient for clinical application and the quality of progenitor cells varies.

“…For example, CD271+ cells in the MSCs isolated from deciduous dental pulp are unable to differentiate into osteogenic, chondrogenic and adipogenic lineages, but the fraction of CD271-cells can [Mikami et al, 2011]. In umbilical cord blood, endothelial progenitor cells also express CD271 [Paprocka et al, 2011] and that could be the reason of the limited differentiation exhibited by CB-CD271 cells in this study.…”

“…However, we did not test this feature in the present study. Similarly to the herein examined TAg cells, SV40 T-antigen immortalized endothelial progenitor cells (EPCs) from human cord blood were shown to be strongly positive for CD146 [28], as well as hTERT immortalized EPCs [29]. A recent study describes CD146 expression in mouse p19 ARF-/- , which show longer tumor latency as p53 -/- mice [30].…”

“…Two cell lines, namely HEPC-CB.1 (55.1) and HEPC-CB.2 (55.2), characterised previously by Paprocka and coworkers [18], were isolated from human cord blood. The EP 55.1 cells were cultured in OptiMEM medium (Invitrogen) supplemented with 3 % FBS, 2 mM glutamine and antibiotics (100 U/ml penicillin, 100 lg/ml streptomycin), under a humidified atmosphere of 5 % CO 2 in air at 37°C.…”

“…Paprocka and co-workers described two cell lines of human endothelial cells, isolated from the cord blood and successfully immortalised. These cell lines, named initially HEPC-CB.1 and HEPC.CB-2, express a panel of stem and endothelial cell markers but not the markers of mature, differentiated endothelial cells [18]. EPCs are involved in ischaemic regeneration by angiogenesis and myogenesis modulation and remodelling of the ischaemic cardiac tissue [16,19].…”
