CD1d Ligands: The Good, the Bad, and the Ugly

Abstract: The MHC class I-like CD1d glycoprotein is a member of the CD1 family of Ag-presenting molecules and is responsible for the selection of NKT cells. A number of ligands that can be presented by CD1d to NKT or other CD1d-restricted T cells have been identified. These include glycolipids from a marine sponge, bacterial glycolipids, normal endogenous glycolipids, tumor-derived phospholipids and glycolipids, and nonlipidic molecules. The presentation of many of these molecules can have immunopotentiating effects, su… Show more

“…It is well documented that glycolipids, including GPI's from T. brucei, can be presented in the context of CD1d, expressed on APCs, resulting in the activation of CD1d-restricted T cells (27,48,49). Therefore, the possible involvement of the CD1d-mediated pathways in the protective effects observed in GPI-based treated/infected animals was envisaged and evaluated using CD1d Ϫ/Ϫ mice.…”

“…20 Those CD1d-restricted NKT cells that express another TCRa chain are simply designated 'non-canonical' or 'variant'. 20 The presentation of antigens by CD1d molecules stimulates NKT cells which can mediate perforin-dependent cytotoxicity. The upregulation of CD40L on activated NKT cells leads to the production of interleukin-12 (IL-12) by dendritic cells (DC), 21 and IL-12 activates NKT cells to produce interferon-c (IFN-c), which, in turn, stimulates NK cells and CD8 + cytotoxic T Lymphocytes (CTL).…”

“…Stimulation of type I NKT cells results in the rapid production of cytokines, including IFN-␥ and IL-4. Although a number of endogenous and pathogen-derived ligands capable of activating type I NKT cells have been described (8), the prototypical ligand remains ␣-galactosylceramide (␣-GalCer) that was originally isolated from the marine sponge Agelas mauritianus (9). ␣-GalCer-CD1d tetramers can be used to specifically identify type I NKT cells in mice and humans (10).…”
