Celastrus paniculatus Willd. mitigates t-BHP induced oxidative and apoptotic damage in C2C12 murine muscle cells

Abstract: Identification, exploration and scientific validation of antioxidant rich herbal extracts to mitigate the radical induced cell damage provide new insights in the field of ayurvedic research/therapies. In the present study, we evaluated the anti-oxidant and anti-apoptotic potential of Celastrus paniculatus seed extract (CPSE) against tertiary butyl hydroperoxide (t-BHP) induced mice muscle cell damage. The extract at a dose of 50 µg/ml protected the cells up to 70 % as evidenced by the 3-(4,5-dimethylthiazol-2-… Show more

“…Lead, a ubiquitous heavy metal and a known toxicant also produces adverse effects on the brain via increased production of reactive oxygen species (ROS) and causes oxidative stress. 22 This, in turn, results in oxidative stress at the cellular level causing a reduction in the sarcolemmal calcium pump and Na + K + ATPase intracellular Ca 2+ overload. Lead can substitute for calcium ions (Ca 2+ ) and picomolar concentrations of Pb 2+ can replace micromolar concentrations of Ca 2+ in a protein kinase C (PKC) enzyme assay, a calcium-dependent process.…”

“…Pre‐treatment with the extract also controlled the responsible markers including superoxide dismutase and catalase amount facilitated by protein expression. Anti‐apoptotic responses by regulation of cytochrome‐C and thermal shock protein‐70 expression were seen that resulted in 43% inhibition of DNA distortion in muscle cells against t‐BHP (Kumar et al, 2015).…”

“…The extract confirmed its potential via restoration of mitochondrial membrane and diminished the production of reactive oxygen species and decreased lipid per oxidation.Pre-treatment with the extract also controlled the responsible markers including superoxide dismutase and catalase amount facilitated by protein expression. “…ROS production was measured by uorogenic dye 2',7'-dichlorodihydro uorescein diacetate (DCFH 2DA ), which is oxidized by intracellular ROS, estimated according to Kumar et al (2015). The SH-SY5Y cells were seeded in 24-well plates (5 x 10 4 cells/ml) and exposed to Fb1 (150μM) following pretreatment with apocynin (100μM) and incubated for 24h.…”

“…Mitochondrial membrane potential (MMP) was determined uorometrically using cationic uorescent dye rhodamine 123, estimated according to Kumar et al (2015) which enters the mitochondrial matrix resulting in photo luminescent quenching which depends on MMP. The SH-SY5Y cells were seeded in 24well plates (5 x 10 4 cells/ml) and exposed to Fb1 (150μM) following pretreatment with apocynin (100μM) and incubated for 24h.…”
