Cell proteins bind to multiple sites within the 5' untranslated region of poliovirus RNA.

Abstract: The 5' noncoding region of poliovirus RNA contains sequences necessary for translation and replication. These functions are probably carried out by recognition of poliovirus RNA by cellular and/or viral proteins. Using a mobility-shift electrophoresis assay and 1,10-phenanthroline/Cu+ footprinting, we demonstrate specific binding of cytoplasmic factors with a sequence from nucleotides 510-629 within the 5' untranslated region (UTR). Complex formation was also observed with a second sequence (nucleotides 97-182… Show more

“…Most probably, ~57 represents only one of several cellular factors implicated in the specific ribosome-RNA interaction, since binding of proteins to multiple sites of the 5 ' UTR of poliovirus has been observed [7]. The identity of ~57 is not clear.…”

“…Binding of cellular proteins to multiple sites [7], and interaction of a 52 kDa protein with a specific region of this internal entry site of poliovirus RNA [8] have been reported. Very recently, binding of a 58 kDa protein from mouse ascites cells to the 5 ' -UTR of encephalomyocarditis virus (EMCV) has been observed upon UV-crosslinking [9].…”

“…PTB and eIF-2␣ have been shown to interact with PV RNA encompassing the putative SLII (14,25). Inefficient recruitment of factors essential for IRES function might explain the growth-restricted phenotype on MFHF and BNL CL.2 cells.…”

“…Cells were incubated at 37°C and 5% CO 2 for 3 days or until cytopathic effect (CPE) was observed and then were thrice frozen and thawed. Viral progeny were clarified by centrifugation at 3,000 ϫ g, and 500 l of supernatant was added to 10 6 HeLa cells and incubated until complete CPE was observed.…”
