Cells of extraembryonic mesodermal origin confer human dystrophin in the Mdx model of duchenne muscular dystrophy

Abstract: Duchenne muscular dystrophy is an X-linked recessive genetic disease characterized by severe skeletal muscular degeneration. The placenta is considered to be a promising candidate cell source for cellular therapeutics because it contains a large number of cells and heterogenous cell populations with myogenic potentials. We analyzed the myogenic potential of cells obtained from six parts of the placenta, that is, umbilical cord, amniotic epithelium, amniotic mesoderm, chorionic plate, villous chorion, and decid… Show more

“…In this study, we only expanded MSCs from placental tissues from uncomplicated pregnancies. However, we also revealed different characteristics among CP-MSCs, CV-MSCs and DB-MSCs, supporting the idea of heterogeneity and tissue-specificity among placental MSCs for cell properties and proliferative capacities13424454. We have started to expand and characterize placental MSCs from abnormal conditions (e.g.…”

“…These explants produced adequate numbers of cells with high proliferative potency. Numerous methods have been used to isolate/expand MSCs from placental tissue, including enzymatic digestion of tissues to harvest MSCs as a single-cell suspension for further cell expansion4344, or by seeding tissue fragments, as in the current study. Contamination between maternal- and foetal-origin MSCs remains a problem with expansion of MSCs from placental tissues4546, but we were able to generate pure cells without contamination.…”

“…Cells-Endometrium (UtE) (29), placental artery endothelium (PAE) (30), and amnion (AM) (31,32) were independently established. UtE, AM, and MRC5 cells were maintained in the POWEREDBY10 medium (MED SHIROTORI CO., Ltd.).…”

“…They exhibit multipotency, that is, adipogenic, osteogenic, and chondrogenic differentiation in vitro [10]. They also show myogenic differentiation in vitro like mesenchymal stem cells derived from the bone marrow, menstrual blood, and placenta [6, 8, 13, 21–23]. These mesenchymal stromal cells can be used for therapeutic agents or delivery vehicles to patients with graft-versus-host disease, ischemic heart disease, and lysosomal storage disorders.…”
