Cellular imaging in drug discovery

Abstract: Traditional screening paradigms often focus on single targets. To facilitate drug discovery in the more complex physiological environment of a cell or organism, powerful cellular imaging systems have been developed. The emergence of these detection technologies allows the quantitative analysis of cellular events and visualization of relevant cellular phenotypes. Cellular imaging facilitates the integration of complex biology into the screening process, and addresses both high-content and high-throughput needs.… Show more

“…1 The search for chemical entities modulating important cellular phenotypes, such as proliferation, differentiation, and survival, can now be conducted in biologically relevant cellular systems. In contrast to single-readout assays, high-content screening (HCS) permits the parallel measurement of an array of parameters (high content), extensively characterizing the cellular phenotype.…”

“…The combination of validated immunofluorescent assays, availability of automated microscopy, and advances in image analysis software led to increased exploitation of this technology in a variety of settings ranging from cellular signaling pathways to drug discovery (6,7). Recently, others have shown the advantages of HCI by revealing complex interactions between experimental molecules and cellular populations using several variables (8 -10).…”

“…First, most experiments continue to focus treatment effects on an entire population of cells and how one or two variables are affected in individual cells (6,7,9). In these experiments, the population of cells is viewed as giving a singular response rather than a combination of several subpopulations, all potentially responding differently to treatment.…”

“…1 The complex intracellular responses involved in drug-induced efficacy or cytotoxicity can be observed in organ-specific cells by HCS. 2,3 This capability of HCS allows the acquisition of cellular information in more detail and provides a useful basis for making better decisions on progressing compounds.…”
