Cellular models of Batten disease

Abstract: The Neuronal Ceroid Lipofuscinoses (NCL), otherwise known as Batten disease, are a group of neurodegenerative diseases caused by mutations in 13 known genes. All except one NCL is autosomal recessive in inheritance, with similar aetiology and characterised by the accumulation of autofluorescent storage material in the lysosomes of cells. Age of onset and the rate of progression vary between the NCLs. They are collectively one of the most common lysosomal storage diseases, but the enigma remains of how genetica… Show more

“…Many animal and mammalian cell models have been used to study the pathological mechanisms underlying the NCLs [7,8]. However, these systems present some limitations, such as the costs associated with maintaining small experimental animals (e.g., mice) and the limited number of phenotypes that can be screened in immortalized or patient-derived human cells.…”

“…Cellular models of NCLs are commonly used to understand cellular biology (Minnis et al., 2019), and as platforms for therapeutic efficacy studies (Gavin et al., 2013; Kim et al., 2012; Linterman et al., 2011). In this study we used mass spectrometry (MS) based secretome profiling to characterise disease specific changes in primary cortical cell cultures of neurons, microglia, and astrocytes derived from the Cln6 nclf mouse.…”

“…Yeast provide useful model systems for many human diseases due their genetic tractability. We have developed yeast models to understand the neuronal ceroid lipofuscinoses (NCL), also known as Batten disease, a group of rare childhood inherited neurodegenerative disorders 1 . Most are inherited in an autosomal recessive manner, with mutations in 13 known human genes 2 , some of which are conserved in yeast.…”
