Cerebral mTOR signal and pro-inflammatory cytokines in Alzheimer’s disease rats

Abstract: As a part of Alzheimer’s disease (AD) development the mammalian target of rapamycin (mTOR) has been reported to play a crucial role in regulating cognition and can be used as a neuronal marker. Neuro-inflammation is also a cause of the pathophysiological process in AD. Thus, we examined the protein expression levels of mTOR and its downstream pathways as well as pro-inflammatory cytokines (PICs) in the brain of AD rats. We further examined the effects of blocking mTOR on PICs, namely IL-1β, IL-6 and TNF-α. Our… Show more

“…Following water maze testing, blood and serum were acquired. Routine indices and cytokines (Wang et al, 2016) were measured and the brains of the animals dissected. Four brains from each group were fixed in 4% paraformaldehyde solution and prepared as paraffin sections.…”

“…Following the MWM, the blood and serum were acquired, and the brains of the animals were dissected. Routine index and cytokines ( Wang et al, 2016 ), including the production of cytokines interleukins [(1L)-1α, 1L-2, 1L-8, 1L-10, 1L-11, IL-12], tumor necrosis factor (TNF)-γ, TNF-α, vascular endothelial growth factor (VGEF), human macrophage inflammatory protein-1α (MIP-α), and macrophage colony-stimulating factor (M-CSF), activities of malondialdehyde (MDA), total superoxide dismutase (T-SOD), catalase (CAT), glutathione reductase (GSH-Px), and levels of acetylcholine (ACh), acetylcholinesterase (AChE), and Na + /K + -ATPase, and some monoamine neurotransmitters, were measured.…”

“…After completion of antibody injection, drugs were given. The following procedures were performed as described in our previous publication (22). Animals were cannulated with an L-shaped stainless steel cannula aimed at the lateral ventricle (coordinates: 3.7 mm posterior to the bregma, 4.1 mm lateral to the midline, and 3.5 mm under the dura).…”
