Changes in Amines and Biosynthetic Enzyme Activities in p-Fluorophenylalanine Resistant and Wild Type Tobacco Cell Cultures

Abstract: The levels of free amines and the activities of their biosynthetic enzymes were measured in a p-fluorophenylalanine resistant Nicotiana tabacum L. cv Xanthi cell line (TX4) which accumulates high levels of cinnamoylamides, and a wild type cell line (TX1). Putrescine in TXI and spermidine in TXI and TX4 increased 4-fold by day 4 but declined by day 8 of the culture period. “…ODC may have a similar function, as it was also increased in TX4 cells, but this pattern may be complicated by the involvement of ODC in other pathways. Recently, Walker et al (22) detected distinctly higher arginine decarboxylase (ADC) activity in TX4 cells and concluded that putrescine may be formed mainly by ADC rather than by ODC. We therefore also measured ADC activity on several days during the kinetic study.…”

“…Though the biochemical comparison of parent and variant lines provided some clues as to the biochemical requirements of overproducing lines, we did not analyze the system at the molecular level. One reason for this was the finding that not only the hydroxycinnamoyl putrescine pathway but also other enzyme activities such as tyrosine decarboxylase (WALKER et al, 1986) were altered in the pfluorophenylalanine resistant cell line.
