BackgroundTomato yellow leaf curl virus (TYLCV) is a member of the family Geminiviridae, genus Begomovirus. The virus is a widespread plant virus that causes important economic losses in tomatoes. Genetic engineering strategies have increasingly been adopted to improve the resistance of tomatoes to TYLCV.ResultsIn this study, a proteomic approach was used to investigate the molecular mechanisms involved in tomato leaf defense against TYLCV infection. Proteins extracted from leaves of resistant tomato cultivar ‘Zheza-301’ and susceptible cultivar ‘Jinpeng-1’ after TYLCV infection were analyzed using two-dimensional gel electrophoresis. Eighty-six differentially expressed proteins were identified and classified into seven groups based on their functions. For several of the proteins, including CDC48, CHI and HSC70, expression patterns measured using quantitative real-time PCR differed from the results of the proteomic analysis. A putative interaction network between tomato leaves and TYLCV infection provides us with important information about the cellular activities that are involved in the response to TYLCV infection.ConclusionsWe conducted a comparative proteomic study of TYLCV infection in resistant and susceptible tomato cultivars. The proteins identified in our work show a variety of functions and expression patterns in the process of tomato–TYLCV interaction, and these results contribute to our understanding of the mechanism underlying TYLCV resistance in tomatoes at the protein level.Electronic supplementary materialThe online version of this article (doi:10.1186/s12870-016-0819-z) contains supplementary material, which is available to authorized users.

Tobacco mosaic virus (TMV) is a widespread plant virus from the genus Tobamovirus that affects tobacco and tomato plants causing a pathology characterised by cell breakage and disorganisation in plant leaves and fruits. In this study we undertook a proteomic approach to investigate the molecular and biochemical mechanisms potentially involved in tomato fruit defence against the viral infection. The comparison of 2-D gels from control and TMV-infected but asymptomatic tomato fruits revealed changes in several proteins. The differential expression of peptidases, endoglucanase, chitinase and proteins participating in the ascorbate-glutathione cycle in infected fruits suggests that pathogenesis-related proteins and antioxidant enzymes may play a role in the protection against TMV infection. TMV coat protein appeared as a prominent spot in 2-D gels from TMV-infected asymptomatic fruits. A Triton X-114 phase-partitioning step of tomato protein extracts favoured the solubilisation of TMV coat protein and the enrichment of two aminopeptidases not present in control fruits. PMF and MS/MS data of the 2-D gel-isolated TMV coat protein is proposed as a powerful analysis method for the simultaneous tobamovirus detection, species determination and strain differentiation in virus-infected fruit commodities.

Accumulation of extracellular chitinases in Brassica napus plants infected with Turnip yellow mosaic virus (TYMV) and fungal pathogen Leptosphaeria maculans was studied in both compatible and incompatible interaction. Analysis of apoplast fluid by means of non-denaturing anodic and cathodic PAGE followed by in-gel detection of chitinase activity revealed a number of chitinase isozymes. TYMV induced 8 acidic and 4 basic isozymes in a systemic way. Except for one acidic and one basic isozyme, all other chitinases were also constitutively present in low amounts in mock inoculated control. In TYMV systemically infected plants, chitinases were detected in leaves expressing symptoms as well as in symptomless ones. Both virulent and avirulent L. maculans isolates induced production of chitinase isozymes in cotyledons in a time dependent manner. Some of them were present in plants constitutively and their content increased after inoculation. Three of five acidic and two of three basic isozymes responded to L. maculans infection. Chitinases started to accumulate before symptom appearance. First two acidic isozymes were detected 24 h after inoculation. The difference between compatible and incompatibe interaction reflected two basic isozymes.Additional key words: oilseed rape, plant defence, PR-proteins, Turnip yellow mosaic virus.
