First published September 8, 2011; doi:10.1152/ajpgi.00061.2011.-Increased cholangiocyte growth is critical for the maintenance of biliary mass during liver injury by bile duct ligation (BDL). Circulating levels of testosterone decline following castration and during cholestasis. Cholangiocytes secrete sex hormones sustaining cholangiocyte growth by autocrine mechanisms. We tested the hypothesis that testosterone is an autocrine trophic factor stimulating biliary growth. The expression of androgen receptor (AR) was determined in liver sections, male cholangiocytes, and cholangiocyte cultures [normal rat intrahepatic cholangiocyte cultures (NRICC)]. Normal or BDL (immediately after surgery) rats were treated with testosterone or antitestosterone antibody or underwent surgical castration (followed by administration of testosterone) for 1 wk. We evaluated testosterone serum levels; intrahepatic bile duct mass (IBDM) in liver sections of female and male rats following the administration of testosterone; and secretin-stimulated cAMP levels and bile secretion. We evaluated the expression of 17␤-hydroxysteroid dehydrogenase 3 (17␤-HSD3, the enzyme regulating testosterone synthesis) in cholangiocytes. We evaluated the effect of testosterone on the proliferation of NRICC in the absence/presence of flutamide (AR antagonist) and antitestosterone antibody and the expression of 17␤-HSD3. Proliferation of NRICC was evaluated following stable knock down of 17␤-HSD3. We found that cholangiocytes and NRICC expressed AR. Testosterone serum levels decreased in castrated rats (prevented by the administration of testosterone) and rats receiving antitestosterone antibody. Castration decreased IBDM and secretinstimulated cAMP levels and ductal secretion of BDL rats. Steroid hormones are important players to regulate adult neurogenesis in the dentate gyrus of hippocampus, but their involvement in the regulation of the same phenomenon in the subventricular zone (SVZ) of the lateral ventricles is not completely understood. Here, in male rats, we tested the existence of activational effects of Testosterone (T) on cell proliferation in the adult SVZ. To this aim, three groups of male rats: castrated, castrated and treated with T, and controls were treated with BrdU and killed after 24 hours. The density of BrdU labeled cells was significantly lower in castrated animals in comparison to the other two groups, thus supporting a direct correlation between SVZ proliferation and levels of circulating T. To clarify whether this effect is purely androgen-dependent, or mediated by the T metabolites, estradiol (E 2 ) and dihydrotestosterone (DHT), we evaluated SVZ proliferation in castrated males treated with E 2 , DHT and E 2 +DHT, in comparison to T-and vehicle-treated animals, and sham-operated controls. The stereological analysis demonstrated that E 2 and T, but not DHT, increase proliferation in the SVZ of adult male rats. Quantitative evaluation of cells expressing the endogenous marker of cell proliferation PHH3, or the marker of highly dividing SVZ progenitors Mash1, indicated the effect of T/E 2 is mostly restricted to SVZ proliferating progenitors. The same experimental protocol was repeated on ovariectomized female rats treated with E 2 or T. In this case, no statistically significant difference was found among groups. Overall, our results clearly show that the gonadal hormones T and E 2 represent important mediators of cell proliferation in the adult SVZ. Moreover, we show that such effect is restricted to males, supporting adult neurogenesis in rats is a process differentially modulated in the two sexes.

The pesticides are one of the most potentially harmful chemicals liberated in the environment in an unplanned manner. Dimethoate is widely used as a potent pesticide in many countries and has been shown to produce some adverse health effects. In the present study, we investigated the effect of dimethoate (an organophosphate insecticide) on the reproductive system and fertility of male rats and the possible mechanism underlining its action. Twenty four adult Wistar male rats were divided into 4 groups of 6 animals per group and force-fed with 0, 3.66, 5.50 and 11 mg kg −1 body weight of dimethoate for 90 days. At 80 days of treatment, each males was allows to mate with two primiparous cyclic females. The results showed a significant decrease (p<0.05) of body weight gain in rats gavaged with dimethoate compared to control. In addition, this insecticide at the doses of 3.66, 5.5 and 11 mg kg −1 caused a significant (p<0.05) increase of the relative weight of epididymis. Dimethoate at its highest dose caused a decrease in the relative weights of testes and ventral prostate. The doses 3.66, 5.50 and 11.00 mg kg −1 body weight of dimethoate caused a significant decrease (p<0.05) of the sperm concentration and motility. The level of protein and cholesterol in the serum and testes as well as the activity of prostatic acid phosphatase decreased significantly (p<0.05) in rats treated with dimethoate compared to control. Fertility, gestation and parturition indices as well as the litter size decreased significantly in animals treated with 5.50 and 11.00 mg kg −1 body weight of dimethoate compared to control. The testicular histology of animals treated with high doses of dimethoate generally showed span of Sertoli cells destruction and disorganization of the germinal epithelium. The lumen of seminiferous tubules of treated rats was generally poor in sperm. The results of this study confirmed that dimethoate seriously deteriorate the male reproductive system resulting in decreased fertility.
