Changes in the chloroplast proteome following water deficit and subsequent watering in a high- and a low-drought-tolerant genotype of Festuca arundinacea

Abstract: Festuca arundinacea is one of the most drought-tolerant species within the Lolium-Festuca complex and was used as a model for research aimed at identifying the chloroplast components involved in the proteomic response for drought stress in forage grasses. Individual F. arundinacea genotypes with contrasting levels of drought tolerance, the high-drought-tolerant (HDT) and the low-drought-tolerant (LDT) genotypes, were selected for comparative physiological and proteomic work. Measurements of water uptake, chlor… Show more

“…It is noteworthy that the number of genes up-or down-regulated during the whole priming process (409) was by far higher than the number of genes modified during soaking (74) and drying (40). This leads us to hypothesize that general priming is a more complex process than the simple sum of soaking and drying phase.…”

“…Spot detection and image analyses (normalization, spot matching, protein accumulation analyses) were performed with Image Master 2-D Platinum 6.0 software (GE Healthcare, UK) and the abundance of each protein spot was normalized as a relative volume (% Vol) as described earlier by Kosmala et al [40]. Six images representing three independent biological replicates for each experimental variant, were used.…”

“…In contrast, Kosmala et al. () observed a significant compromise to the photochemical efficiency of PS II in a drought‐sensitive tall fescue cultivar following a short (6 days) mild drought treatment, indicating the significant variability that exists within grass species.…”

“…This method was shown earlier to be useful to extract the proteins from different species and its use resulted in high-quality 2-D maps with proteins derived from different cell components, including membranes, chloroplasts, mitochondria and cytoplasm (Kosmala et al 2009, 2012; Winiarczyk and Kosmala 2009; Bocian et al 2011). Briefly, the powdered tissue was homogenised with 500 μl of extraction buffer (0.7 M sucrose, 0.5 M Tris, 30 mM HCl, 50 mM EDTA, 2 % DTT, 0.1 M KCl).…”
