Changes in wall teichoic acid during the rod-sphere transition of Bacillus subtilis 168

Abstract: Wall teichoic acid (WTA) is essential for the growth of Bacillus subtilis 168. To clarify the function of this polymer, the WTAs of strains 168, 104 rodBl, and 113 tagFl (rodCl) grown at 32 and 42°C were characterized. At the restrictive temperature, the rodBI and tagFl (rodCl) mutants undergo a rod-to-sphere transition that is correlated with changes in the WTA content of the cell wall. The amount of WTA decreased 33% in strain 104 rodBl and 84% in strain 113 tagFl (rodCl) when they were grown at the restrict… Show more

“…Analyses also point toward a role of the TAs in cell division and elongation of Bacillus subtilis 168 (221). WTA plays a role in cell elongation in certain rod-shaped bacteria, as was shown by mutation of WTA, which resulted in a round bacterial morphology instead of a rod in B. subtilis (228). LTA is synthesized at the site of cell division in B. subtilis and is believed to be involved in cell division (229,230).…”

“…Growth of a tagF1 (rodC1) strain with a defect in the Gro-P transferase TagF (Fig. 4) (399) at this temperature gave WTA with chains approximately 8 units long, in contrast to chain lengths of approximately 53 residues when the strain was grown at the permissive temperature (393). This strain contained only 16% of its WTA when grown at the restrictive temperature.…”

“…After translocation across the cytoplasmic membrane, the TA is covalently linked to PG. One of the enzymes involved in the precursor synthesis, TagF, is essential in B. subtilis, and thermosensitive mutants display a spherical phenotype (149)(150)(151). Two other enzymes, TagG and TagH, are thought to act together as a translocase for the lipid-linked TA precursor (105).…”
