The brown planthopper (BPH) is the most devastating insect pest of rice. The rice gene BPH15 confers resistance to BPH. MicroRNAs (miRNAs) regulate a spectrum of development and defense response processes in plants. In this study, we analyzed six miRNA profiles of a BPH15 introgression line (P15) and a susceptible recipient line (PC) at three time points (0 h, 6 h and 48 h) after BPH attack, and identified 464 known miRNAs and 183 potential novel miRNAs. Before the BPH feeding, we identified 23 miRNAs differentially expressed in P15 and PC. We speculated that the resistant plant is in a priming state by the regulation of miRNAs. After the BPH feeding, 104 miRNAs were found to be expressed differentially in P15 (68 in P15-6/P15-0, 36 in P15-48/P15-0), and 80 miRNAs were found expressed differentially in PC (32 in PC-6/PC-0, 48 in PC-48/PC-0), which illustrated that miRNA expression is activated upon attack. These miRNAs regulate different pathways that contribute to the basal defense and specific resistance of rice to the BPH. Our study provides additional data for scientists to further explore the mechanism of plant defense against insect attack and to find a way for efficient insect control.

Insects have a robust capacity to produce offspring for propagation, and the reproductive events of female insects have been achieved at the molecular and physiological levels via regulatory gene pathways. However, the roles of MicroRNAs (miRNAs) in the reproductive development of the brown planthopper (BPH), Nilaparvata lugens, remain largely unexplored. To understand the roles of miRNAs in reproductive development, miRNAs were identified by Solexa sequencing in short-winged (SW) female adults of BPH. Small RNA libraries derived from three developmental phases (1 day, 3 days, and 5 days after emergence) were constructed and sequenced. We identified 905 miRNAs, including 263 known and 642 novel miRNAs. Among them, a total of 43 miRNAs were differentially expressed in the three developmental phases, and 14,568 putative targets for 43 differentially expressed miRNAs (DEMs) were predicted by TargetScan and miRanda. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis of the predicted miRNA targets illustrated the putative roles for these DEMs in reproduction. The progress events were annotated, including oogenesis, lipid biosynthetic process, and related pathways such as apoptosis, ABC transporters, and amino acid metabolism. Four highly abundant DEMs (miR-9a-5p, miR-34-5p, miR-275-3p, and miR-317-3p) were further screened, and miR-34-5p was confirmed to be involved in the regulation of reproduction. Overexpression of miR-34-5p via injecting its mimics reduced fecundity and decreased Vg expression. Moreover, target genes prediction for miR-34-5p showed they might be involved in 20E signaling cascades, apoptosis, and gonadal development, including hormone receptor 4 (HR4), caspase-1 (Cp-1), and spermatogenesis-associated protein 20 (SPATA20). These findings provide a valuable resource for future studies on the role of miRNAs in BPH reproductive development.

Background: Hair follicles is an appendage from the vertebrate skin epithelium, and arise from the embryonic ectoderm andregenerate cyclically during adult life. Dermal papilla cells (DPCs) is the key dermal component of the hair follicle that directly regulates hair follicle development, growth and regeneration. Recent studies have reported that miRNA plays an important role in regulating hair follicle morphogenesis, proliferation, differentiation and apoptosis of hair follicle stem cells. Results: The miRNAs expression profile of the DPCs from different hair density Rex rabbits shown that 240 differentially expressed of miRNAs were screened (log 2 (HD/LD)｜>1.00 and Q-value≤0.001). Among them, the expression of ocu-miR-205-5p in low hair densities DPCs was higher than that in high hair densities, and it is highly expressed in the skin tissue of Rex rabbits ( P <0.05). ocu-miR-205 could increase cell proliferation and cell apoptosis ratio, change cell cycle process ( P <0.05), affect the genes expression of PI3K/Akt, Wnt, Notch and BMP signaling pathways in DPCs and skin tissue of Rex rabbits, inhibit the protein phosphorylation level of CTNNB1, GSK-3β and the protein expression level of noggin (NOG), promote Akt phosphorylation level ( P <0.05). There was no significant change in primary follicle density ( P >0.05), but the secondary follicle density and total follicle density ( P <0.05) were changed after ocu-miR-205-5p interfered expression, and secondary/primary ratio (S/P) in ocu-miR-205-5p interfered expression group increased at 14 days after injection ( P <0.05). Conclusion: ocu-miR-205 could promote the apoptosis of DPCs, affect PI3K/Akt, Wnt, Notch and BMP signaling pathways genes and proteins expression in DPCs and skin of Rex rabbits, promote the transformation of hair follicles from growth phase to regression and resting phase, and affect hair density of Rex rabbits.
