Characterization and validation of point mutation in Exon 19 of Calcium Channel, voltage-dependent, Alpha-2/Delta subunit 1(CACNA2D1) gene and its relationship with mastitis traits in Sahiwal

Abstract: Calcium channel, voltage-dependent, alpha-2/ delta subunit 1 (CACNA2D1) gene plays an important role in excitation-contraction coupling in muscle-cells, glial cells and neurons. The CACNA2D1 gene may be one of the candidate genes related with some phenotypic traits due to its location in QTLs associated with Somatic cell score (SCS ) and mastitis. A total of 120 Sahiwal cattle were selected to characterize exon 19 of CACNA2D1 gene to identify polymorphism and its association with mastitis susceptibility/resist… Show more

“…As described earlier, the ATP synthase mitochondrial F1 complex assembly factor 1 (ATPAF1) and lymphoid enhancer-binding factor-1 (LEF1) gene were identified as candidate gene under selection between Tharparkar and Gir cattle. The variants of CACNA2D1 gene were also found to be associated with somatic cell score (SCS) and mastitis resistance/susceptibility in Sahiwal cattle (Magotra et al, 2016) of India. The sulfatase modifying factor 1 (SUMF1) gene on chromosome 22 was also found within the selection signature region and this gene is related with the metabolism of lipids and lipoproteins.…”

“…Mastitis, characterized by inflammation of the mammary glands, can result from trauma or microbial invasion (Cheng & Han, 2020). In the dairy industry, it presents a significant global challenge, causing economic losses through reduced milk production (Hamadani et al, 2013;Magotra et al, 2016Magotra et al, , 2017Oliveira et al, 2013), compromised conception rates (Cheng & Han, 2020), animal welfare concerns, and financial burdens (Ruegg, 2017). Clinical mastitis, manifesting as physical, chemical, and bacteriological alterations in milk (Reis et al, 2013), alongside pathological changes in udder tissue, signifies a symptomatic manifestation of this condition (Kamaldeep et al, 2019;Magotra et al, 2020).…”

“…Efforts to enhance mastitis resistance involve genetic selection methods based on somatic cell count (SCC) as a surrogate trait (Magotra et al, 2018; Miglior et al, 2005). Yet, direct breeding for mastitis resistance faces challenges like low heritability, potential immune system bias, and genetic complexity (Gaddis et al, 2014; Koeck et al, 2012; Thompson‐Crispi et al, 2012).…”

“…Globally, India possesses world's largest bovine population which is about 14.7% (Magotra et al, 2016).The total cattle population is 190.90 millions out of which indigenous cattle population is 151.17 millions and exotic/ crossbred cattle population is 39.73 millions (19 th Livestock census, 2012). Low productivity of native breeds and production related diseases are major concerns of the livestock industry of India..…”
