Characterization of a tandem repeat polymorphism in Rickettsia strains

Abstract: Mediterranean spotted fever (MSF) is a tick-borne rickettsiosis caused by 'Rickettsia conorii complex' strains. In Portugal, R. conorii and Israeli tick typhus (ITT) are the aetiological agents of this disease. A novel 65 bp tandem repeat was identified by the analysis of the R. conorii Malish 7 whole genome sequence with an appropriate algorithm for searching for repeated sequences. The variable number tandem repeat (VNTR) was named VNTR Rc-65 and this locus was amplified by PCR and sequenced in order to char… Show more

“…The resulting electropherograms from the VNTR typing assay are in agreement with amplicon sizes assessed by agarose gel electrophoresis and with the sequencing data previously obtained for each rickettsiae 4 . Nevertheless, minor differences (1 bp [base pairs]) in the highest pick can be observed because 3′ adenine addition by the DNA polymerase.…”

“…Such a variability makes the dksA-xerC the most discriminatory of the three spacers, with 42 genotypes identified. In a recent work, Vitorino et al proposed that PCR amplification of VNTRs within the dksA-xerC might serve to identify rickettsial isolates [53]. However, these authors only took into account the size of the VNTRs, not their sequence variability.…”
