Characterization of Additional Casein Kinase I Sites in the C‐Terminal “Tail” Region of Chicken and Rat Neurofilament‐M

Abstract: Abstract:In previous studies we have identified Ser 502, Ser528, and Ser534 as target sites in chicken neurofilament middle molecular mass protein (N F-M) for casein kinase I (CKI) in vitro and have shown that these sites are also phosphorylated in vivo. We now make use of a combination of molecular biological and protein chemical techniques to show that two additional in vivo phosphorylation sites in chicken NF-M, Ser464 and Ser471, can also be phosphorylated by CKI in vitro. These two sites are conserved in … Show more

“…We next compared the influence of overexpression of GSK3b and CK1, because the majority of NF-H consensus sites for these kinases exist within the distal-most 187 amino acids (Chen et al, 2000;Hollander and Bennett, 1992;Hollander et al, 1996;Sasaki et al, 2002;Shaw et al, 1997), on bundling of GFP-NF-H and GFP-NF-HD187 (Fig. 8E).…”

“…Ser473 in the NF‐L E‐segment is phosphorylated in vivo (Xu et al, 1990) by casein kinase II (Nakamura et al, 1999). The chicken NF‐M E‐segment is phosphorylated by NF‐associated casein kinase I (Hollander and Bennett, 1992; Hollander et al, 1996; Shaw et al, 1997). We have shown that Ser493 in the NF‐H E‐segment is phosphorylated by glycogen synthase kinase 3β (GSK3β) in vitro and invivo (Sasaki et al, 2002).…”

“…We have identified five such sites in the acidic amino‐terminal portion of the chicken NF‐M tail that are also phosphorylated in vivo (Ser 464 , Ser 471 , Ser 502 , Ser 528 , and Ser 536 ; Fig. 1A) (Hollander et al, 1996 ; Shaw et al, 1997). At least three of these are conserved in the corresponding mammalian sequences (Shaw et al, 1997).…”

“…1A) (Hollander et al, 1996 ; Shaw et al, 1997). At least three of these are conserved in the corresponding mammalian sequences (Shaw et al, 1997). These sites all share a casein kinase I (CKI) consensus motif (D/E 2‐4 X 0‐2 XS/T), and they can be phosphorylated in vitro by pure CKI (Hollander et al, 1996 ; Shaw et al, 1997).…”
