Characterization of KIR + NKG2A + Eomes− NK‐like CD8+ T cells and their decline with age in healthy individuals

Abstract: Background: KIR+NKG2A + Eomes+ CD8+ T cells, which are preferentially found with a T EMRA (CD45RA + CCR7−) phenotype while having the capacity to rapidly produce IFN-γ in response to innate stimulation , have been demonstrated to exist in human cord blood and the adult blood circulation. This highly responsive T-cell type was termed NK-like CD8+ T cells due to their capability to act in an innate immune fashion in mice similar to NK cells. However, KIR+NKG2A + CD8+ T cells that are Eomes− represent a small pro… Show more

“…Lastly, we documented that this phenomenon preexists in healthy controls. Indeed, frequency of Innate E(+) CD8 T-cells was apparently not related to age, corroborating findings from Kasakovski et al obtained with a larger healthy cohort aged from 6 to 84 years ( 81 ). Moreover, the healthy control Innate E(+) CD8 T-cell population appeared more enriched in senescent cells compared to its conventional memory counterpart, whose senescent phenotype increased with age.…”

“…Unconventional T cells bridge innate and adaptive immunity, which typically exhibits rapid immune responses, including γδ T cells or our previously described KIR + NKG2A + Eomes − NK‐like CD8 + T cells (Bagwell, 2021; Kasakovski et al, 2021). Included in these T cells are MAIT cells, which have been indicated to be involved in the immunopathology of autoimmune disease (Toubal et al, 2019).…”

“…Cell surface staining analysis for PD‐1, Tim‐3, CD244, CD25, CD3, CD4, CD8, and CD45 and intranuclear staining analysis for TOX and FoxP3 were performed with a BD FACS Canto flow cytometer (BD Biosciences, San Jose, USA) and Flowjo software (Flowjo LLC, USA) as previously described (Huang et al, 2021; Kasakovski et al, 2021). The antibodies used for this study were purchased from BD Biosciences (San Jose, USA) and eBioscience (San Diego, USA).…”
