Characterization of Laminin Binding Integrin Internalization in Prostate Cancer Cells

Abstract: Laminin binding integrins α6 (CD49f) and α3 (CD49c) are persistently but differentially expressed in prostate cancer (PCa). Integrin internalization is an important determinant of their cell surface expression and function. Using flow cytometry, and first order kinetic modeling, we quantitated the intrinsic internalization rates of integrin subunits in a single cycle of internalization. In PCa cell line DU145, α6 integrin internalized with a rate constant (kactual) of 3.25 min−1, threefold faster than α3 integ… Show more

“…These markers were chosen due to known membranous expression levels (CD49f) and cytoplasmic and nuclear (HMWCK + p63 respectively) positive expression levels in non-neoplastic basal cells of prostate tissue. In PCa, CD49f expression is membranous and aggressive and invasive disease exhibits an intracellular expression pattern [14][15][16][17][18]. It is also associated with poor patient prognosis, reduced survival, and increased metastasis [19][20][21].…”

“…Moreover, laminin binds various integrins receptors (α 1 β 1 , α 2 β 1 , α 3 β 1 , α 6 β 1 , α 7 β 1 , α 10 β 1 , α 6 β 4 , and α v β 8 ) (Alam et al, 2007). Laminin-1, 2, 5, 8, 10, 11 isoforms interact with integrins α 3 β 1 and α 6 β 1 which regulate embryonic development, epithelial regeneration, and wound healing processes, and which also internalize to endosome as well (Das et al, 2017). Laminin binding cell receptors are highly expressed in various cancer cells types.…”
