Characterization of Peptides Bound to the Class I MHC Molecule HLA-A2.1 by Mass Spectrometry

Abstract: Antigens recognized by T cells are expressed as peptides bound to major histocompatibility complex (MHC) molecules. Microcapillary high-performance liquid chromatography-electrospray ionization-tandem mass spectrometry was used to fractionate and sequence subpicomolar amounts of peptides isolated from the MHC molecule HLA-A2.1. Of 200 different species quantitated, eight were sequenced and four were found in cellular proteins. All were nine residues long and shared a distinct structural motif. The sensitivity … Show more

“…Most studies to date used untargeted unbiased LC‐MS 2 detection, which detects large numbers of epitopes presented at higher abundance. Untargeted detection was used for identification of HLA binding motives,35 viral epitopes,36 and tumor mutation‐derived epitopes (neoepitopes) from cancer cell lines37, 38 or primary human tumor material 39, 40. However, this common workflow fails to identify low‐abundant peptides that might still be important for immunotherapy development.…”

“…We infer that other endogenously processed, but wild-typeencoded MHC-dependent peptides are likely present in urine in similar or even lower concentrations although they cannot be detected by today's MS. The urinary concentrations of all MHCdependent peptides may add up to approximately 10 À 10 to 10 À 8 M if one assumes that SIINFEKL represents between 0.1 and 10% of the H2-K b ligands 51,52 . In order for these peptides to qualify as pheromone-like cues that influence behaviour, a mouse must be able to recognize the qualitative difference of the MHC-dependent peptidome in the presence of the plethora of other, much more abundant urinary peptides.…”

“…The Rammensee lab reported the first application of mass spectrometry and Edman degradation for the identifications of pMHC,23 while in 1992, Hunt et al 24. published the first LC‐MS/MS approach for the identification of eluted pMHC.…”
